摘 要:以总黄酮含量为指标,通过考察静态、动态吸附、洗脱试验,评价聚酰胺树脂富集金丝草总黄酮的最优工艺,结果表明:最佳工艺参数为上样液黄酮质量浓度2 mg·m L-1,上样量2 BV,加20%乙醇3 BV冲洗杂质,用60%乙醇3 BV洗脱总黄酮,富集后总黄酮纯度提高至39.10%。因此,采用聚酰胺树脂富集金丝草总黄酮的效果良好,为金丝草黄酮的分离纯化与金丝草总黄酮药理试验奠定基础。The aim of this paper is to optimize purifying technology of total flavonoids from herba, Pogonatherum crinitum (Thunb.) Kunth by polyamide resin, and the static and dynamic adsorption and desorption behaviors of polyamide resin were evaluated for achieving an optimal purification of total flavonoids. The results showed that optimum purifying technology parameters were as follows: sample concentration of 2 mg· mL-1 , sample volume of 2 BV, eluted impurities with 3 BV of 20%ethanol, then eluted with 3 BV of 60% ethanol, and purity of total flavonoids was up to 39.10%. Purification effect of total flavonoids from P. crinitum by polyamide resin was feasible, and this study could provide experimental basis for the separation, purification and pharmacological experiment of P. crinitum.
