摘 要:目的探讨黏着斑激酶(FAK)酪氨酸861磷酸化状态在高血压致左心室肥大发生发展中的作用。方法通过免疫荧光标记方法、共聚焦显微镜观察及蛋白质电泳和蛋白质印迹等方法,检测2、6、12、18月龄自发性高血压大白鼠(SHR)左心室中酪氨酸861磷酸化的FAK(FAKp Tyr861)定位和表达的变化。结果蛋白印迹结果显示,在总蛋白匀浆和膜蛋白匀浆中,2月龄的SHR组和相同月龄的对照Wistar-Kyoto(WKY)大鼠组相比较,FAKp Tyr861的表达无显著差异;但FAKp Tyr861在6、12、18月龄SHR组左心室心肌细胞总蛋白匀浆和膜蛋白匀浆中的表达量较相同月龄的对照组明显减少(P<0.05)。免疫荧光标记显示,FAKp Tyr861在6、12、18月龄SHR左心室心肌细胞中出现表达与定位变化,主要表现为心肌细胞闰盘处聚集的线状荧光较同月龄的WKY减弱。结论在高血压左心室心肌肥大的发生发展过程中,FAKp Tyr861可能参与了心肌细胞信号转导通路的调控。Objective To investigate the role of focal adhesion kinase phosphorylated at tyrosine 861 ( FAKpTyr861 ) in the mechanism of hypertensive left ventricle hypertrophy. Methods Expression and location of FAKpTyr861 were determined by immunolabeling, confoeal micros- copy, protein electrophoresis and Western Blot in cardiac myoeytes of the LV from 2, 6, 12 and 18 months old spontaneously hypertensive rats (SHR). Results Western Blot results showed that there were no significant differences in the FAKpTyr861 expression level in the total protein and the membrane protein of the LV in between the 2 months old SHR and 2 months old WKY rats, while FAKpTyr861 expression significantly reduced in the total protein and the membrane protein of the LV in the 6, 12 and 18 months old SHR, as compared with" the age - matched WKY rats respectively. Immunofluoreseenee labeling showed that FAKpTyr861 showed bright large particle and coloealized with pan - eadherin at the interca- lated disc sites, these fluorescence weakened at the intercalated discs of the cardiac myocytes in 6, 12 and 18 months old SHR, as compared with the age - matched WKY rats respectively. Conclusion The present results suggested that FAKpTyr861 may play a role in regulating signal transduction in the cardiac myocytes of the LV hypertensive hypertrophy.
