地塞米松对哮喘大鼠中性粒细胞模式识别受体基因表达的影响

摘  要:目的探讨核苷酸结合寡聚域(NOD)样受体在哮喘发病机制中的作用。方法采用哮喘大鼠模型分离提纯血中性粒细胞(PMN),实时荧光定量PCR法检测PMN NOD2和NALP1 mRNA的表达。结果哮喘组NOD2 mRNA的表达量(3.11±0.82)显著高于对照组(P<0.01),地塞米松组NOD2 mRNA的表达量(1.30±0.28)显著低于哮喘组(P<0.01)。哮喘组NALP1 mRNA的表达量(0.51±0.21)显著低于对照组(P<0.01),地塞米松组NALP1 mRNA的表达量(0.91±0.26)显著高于哮喘组(P<0.05)。血PMN NOD2和NALP1 mRNA的表达水平呈显著负相关性(n=24,r=-0.74,P<0.01)。结论哮喘大鼠血PMN NOD2 mRNA的表达升高,NALP1 mRNA的表达则相反,地塞米松具有调节模式识别受体的作用。Objective To investigate the potential roles of nucleotide-binding oligomerization domain(NOD)-like recep- tor in the pathogenesis of asthma. Methods Blood polymorphonuclear (PMN) was isolated and purified from rat asthma model, expression levels of NOD2 and NALP1 mRNA were detected by real-time PCR methods. Results Levels of NOD2 mRNA in asthma group(3.11±0.82) was significantly higher than that in control group(P〈0.01), while it was sig- nificantly decreased in dexamethasone group(1.30±0.28) when compared with asthma group(P〈0.01). Whereas, levels of NALP1 mRNA in asthma group(0.51±0.21)was significantly lower than that in control group(P〈0.01), while level of NALP1 mRNA in dexamethasone group(0.91±0.26) was significantly increased when compared with asthma group(P〈 0.05). Moreover, level of NOD2 mRNA was strongly negative correlated with NALP1 mRNA(n=24, r=-0.74, P〈0.01). Con- clusion Expression levels of PMN NOD2 mRNA is elevated at rat asthma, while NALP1 mRNA is decreased, functions of pattern recognition receptors can be regulated by desamethasone.
