黑地黄丸改善5/6肾切除大鼠肾衰模型肾功能的研究

摘　　要：目的:通过观察黑地黄丸对5/6肾切除肾衰大鼠(CRF大鼠)的血清尿素氮(BUN)、肌酐(Scr)水平的影响,探讨黑地黄丸对CRF大鼠肾功能的影响,并寻求最佳剂量。方法:将70只Wistar大鼠随机分为空白组,模型组,黑地黄丸高、中、低剂量组,尿毒清组,洛丁新组,共7组,除空白组外其他各组大鼠均行5/6肾切除术,建立CRF大鼠模型,连续给药12周后,采用酶偶联速率法测定血清尿素及采用碱性苦味酸法测定血清肌酐动脉含量。结果:与模型组比较,黑地黄丸高、中、低剂量组血清BUN、Scr显著降低(P<0.01)。与黑地黄丸低剂量组比较,黑地黄丸高剂量组、尿毒清组血清BUN降低(P<0.05);与黑地黄丸低剂量组比较,黑地黄丸高剂量组、尿毒清组血清Scr显著降低(P<0.01)。结论:黑地黄丸可以明显降低肾衰大鼠的血清BUN、Scr,从而改善肾功能,延缓CRF进程,其中尤以黑地黄丸高剂量组效果最佳。Objective： To survey the effects of black DiHuang pills on renal function of the rats with chronic renal failure（CRF）, find out the optimal dosage by observing its effects on the levels of serum urea and creatinine of CRF rats undergoing 5/6 nephrectomy. Methods： Seventy Wistar rats were divided into seven groups randomly, the blank group, the model group, and high, middle and low dosages groups of black DiHuang pills, NiaoDuQing group and Lotensin group. The rats received 5/6 nephrectomy except the ones in the blank group to establish CRF rat mod- els, the contents of serum urea were detected by the method of enzyme coupling ratio and the contents of serum crea- tinine detected by basic picric acid method after the rats were medicating for 12 weeks. Results： Compared with the model group, the levels of serum BUN and Scr decreased in high, middle and low dosages groups of black DiHuang pills significantly （P〈0.01）. Compared with low dosages group of black DiHuang pills, the levels of serum BUN of high dosage group of black DiHuang pills and NiaoDuQing group lowered （P〈0.05）. Compared with low dosages group of black DiHuang pills, the level of serum Scr in high dosages group of black DiHuang pills and NiaoDuQing group decreased significantly （P〈0.01）. Conclusion： Black DiHuang pills can decrease the levels of serum BUN and Scr of the rats with renal failure obviously, therefore to improve renal function and delay the progress of CRF, espe- cially high dosage of black DiHuang pills.
