摘  要:目的 评价鞘内注射细胞穿膜肽Tat-FynP对炎性痛大鼠的影响.方法 健康雄性SD大鼠80只,6~8周龄,体重180~ 220 g,采用随机数字表法分为5组(n=16):对照组(C组)、炎性痛组(IP组)、短肽FynP组(FynP组)、Tat-FynP组和乱序细胞穿膜肽mTat-FynP组(mTat-FynP组).IP组、FynP组、Tat-FynP组和mTat-FynP组采用右后足趾皮下注射完全弗氏佐剂100μl的方法制备大鼠炎性痛模型,C组给予等容量生理盐水.于造模后1d时,FynP组、Tat-FynP组和mTat-FynP组分别鞘内注射FynP、Tat-FynP、mTat-FynP各15μl.每组取10只大鼠,分别于造模前和造模后1、3、5、7、10、14d时测定机械缩足反应阈(MWT)和热缩足潜伏期(TWL).鞘内给药后1d时每组取6只大鼠,采用Western blot法测定脊髓磷酸化含2B亚基的NMDA受体(p-NR2B)的表达水平.结果 与C组比较,IP组、FynP组、Tat-FynP组和mTat-FynP组造模后各时点MWT降低,TWL缩短,脊髓组织p-NR2B表达上调(P<0.01);与IP组比较,Tat-FynP组造模后3~14d时MWT升高,TWL延长,脊髓组织p-NR2B表达下调(P<0.01),FynP组和mTat-FynP组上述各指标差异无统计学意义(P>0.05).结论 鞘内注射细胞穿膜肽Tat-FynP可减轻大鼠炎性痛.Objective To evaluate the effect of intrathecal cell-penetrating peptide Tat-FynP on inflammatory pain in rats.Methods Eighty healthy male Sprague-Dawley rats,aged 6-8 weeks,weighing 180-220 g,were randomly divided into 5 groups (n=16 each) using a random number table:control group (group C),inflammatory pain group (group IP),group FynP,group Tat-FynP and group mTatFynP.Inflammatory pain was induced by injecting complete Freund's adjuvant 100 μl into the plantar surface of the right hindpaw in IP,FynP,Tat-FynP,and mTat-FynP groups,while the equal volume of normal saline was given in group C.At 1 day after establishment of the model,FynP,Tat-FynP and mTatFynP 15 μl (20 μg/kg) were injected intrathecally in FynP,Tat-FynP,and mTat-FynP groups,respectively.Ten rats in each group were randomly selected,and the mechanical paw withdrawal threshold (MWT) and thermal paw withdrawal latency (TWL) were measured before establishment of the model and at 1,3,5,7,10 and 14 days after establishment of the model.The left 6 rats were selected from each group at 1 day after intrathecal administration,and the lumbar enlargement segments of the spinal cord were removed for determination of phosphorylated 2B subunit-containing NMDA receptor (p-NR2B) expression by Western blot.Results Compared with group C,the MWT was significantly decreased,and the TWL was significantly shortened at each time point after establishment of the model,and the expression of pNR2B was significantly up-regulated in IP,FynP,Tat-FynP and mTat-FynP groups (P〈0.01).Compared with group IP,the MWT was significantly increased,and the TWL was significantly prolonged on 3-14 days after establishment of the model,and the expression of p-NR2B was significantly down-regulated in group Tat-FynP (P〈0.01),and no significant change was found in the parameters mentioned above in FynP and mTat-FynP groups (P〉0.05).Conclusion Intrathecal cell-penetrating peptide Tat-FynP can reduce inflammatory pain in rats.
