摘  要:目的探讨乳腺癌超声光散射断层成像(US-DOT)与缺氧诱导因子-1α(HIF-1α)的相关性。方法对2007年10月至2010年2月在北京协和医院行手术切除、经病理证实、术前行US-DOT的69例原发性乳腺癌组织切片行HIF-1α、CD34免疫组织化学染色,分析HIF-1α阳性组和阴性组微血管密度(MVD)与US-DOT总血红蛋白浓度(THC)的差异。结果 69例中,HIF-1α阳性病例12例(17.4%),平均THC(274.763±77.661)μmol/L,平均MVD(33.8±10.8)个/0.2 mm2;HIF-1α阴性病例57例(82.6%),平均THC(228.059±65.760)μmol/L,平均MVD(28.4±7.4)个/0.2 mm2;HIF-1α阳性组的MVD(t=2.049,P=0.04)和THC(t=2.167,P=0.034)均显著高于HIF-1α阴性组。结论 HIF-1α可以促进肿瘤血管生成,使肿瘤内血供增加,THC增加。作为反映肿瘤血供的指标,THC可间接反映肿瘤的血管生成活性。Objective To investigate the correlation between ultrasound-guided diffuse optical tomo- graphy (US-DOT) and hypoxia-inducible factor-1α (HIF-1α) of breast cancer. Methods Totally 69 patients with pathologically confirmed breast cancer underwent preoperative conventional breast ultrasonography examina- tions and US-DOT at Peking Union Medical College Hospital From October 2007 to February 2010 were enrolled in this study. After surgery, immunohistochemical staining of HIF-1 α and CD34 were performed, and the differ- (33.8± 10. 8) /0. 2 mm2 respectively. The average THC and MVD of HIF-lα-negative cases were (228. 059 ± 65.760) μmoL/Land (28.4±7.4) /0.2 mm2. MVD (t =2.049, P=0.04) and THC (t=2.167, P= 0. 034 ) of HIF-1 a-positive group were significantly higher than those of HIF-1α-negative group. Conclusions HIF-1 α can promote tumor angiogenesis and thus increase the blood supply and THC. As an indicator of tumor blood supply, THC can indirectly reflect the angiogenie activity of breast cancer.
