摘 要:目的研究肾茶对小鼠血尿酸水平的影响。方法采用腹腔注射次黄嘌呤的方法建立高尿酸血症动物模型,正常小鼠和高尿酸血症模型小鼠以灌胃方式分别接受低、中和高剂量肾茶(一日2 000、4 000及8 000 mg/kg),连续3 d。采用全自动生化分析仪测定小鼠血尿酸浓度,比较各剂量肾茶给药后正常组和模型组小鼠血尿酸水平的变化。结果腹腔注射次黄嘌呤45 min后小鼠血尿酸水平达到峰值。低剂量肾茶对正常小鼠的血尿酸水平无显著影响(P>0.05),中剂量和高剂量肾茶可显著降低正常小鼠的血尿酸水平(P<0.01)。高尿酸血症模型小鼠腹腔注射次黄嘌呤45 min后采集血清标本测定,与不造模对照组相比,模型组小鼠血尿酸水平显著升高(P<0.01),表明造模成功。与模型组比较,低剂量肾茶对高尿酸血症小鼠血尿酸水平无明显影响(P>0.05),中剂量和高剂量肾茶可显著降低高尿酸血症小鼠的血尿酸水平(P<0.01),且具有剂量相关性。结论腹腔注射次黄嘌呤可成功建立高尿酸血症动物模型,肾茶可降低正常小鼠和高尿酸血症模型小鼠的血尿酸水平。Objective To investigate the effect of Clerodendranthus spicatus on serum uric acid level in mice. Methods Intraperitoneal injected hypoxanthine was used to build the mice model of hyperuricemia. The low, middle and high dose of C spicatus (2 000, 4 000 and 8 000 mg/kg/d) were intragastrically administrated to normal mice and hyperuricemia mice respectively, and continuously for 3 days. The automatic chemistry analyzer was used to measure the serum uric acid level. Results The peak of serum uric acid level appeared at 45 min after intraperitoneal injection of hypoxanthine. There was no significant effect on serum uric acid level in both model group and control group when using low dose of C spicatus (P〉0.05). The serum uric acid level in model groups and control group was significantly decreased when using middle dose and high dose of C. spicatus (P〈0.01), which showed dose-dependent correlation. Conclusion The hyperuricemia animal model can be successfully established by intraperitoneal injecting hypoxanthine. C spicatus can reduce the serum uric acid level of normal and hyperuricemia mice.
