摘 要:目的:探讨纯康血脂胶囊对高脂血症大鼠肝损伤的保护作用。方法:40只雄性SD大鼠随机分为空白组、模型组、辛伐他汀组及纯康血脂胶囊组。高脂饲料饲喂3周建立高脂血症动物模型,各组大鼠给予相应的药物灌胃9周,检测血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C),苏木素-伊红(HE)染色观察药物对肝组织病理学改变,半定量RT-PCR法检测肝组织细胞间黏附分子-1(ICAM-1)、血管细胞黏附分子-1(VCAM-1)基因表达变化。结果:与模型组相比,喂饲高脂血症饲料大鼠在连续灌胃给予纯康血脂胶囊后,大鼠血清TC、TG、LDL-C含量分别下降37.47%、55.91%、72.56%,HDL-C含量增加3.34倍。与模型组比较,纯康血脂胶囊组ICAM-1、VCAM-1的基因表达均下调,分别下调19.56%、30.41%(P<0.01);组织病理学显示,纯康血脂胶囊组肝损伤轻于模型组。结论:纯康血脂胶囊可改善高脂血症大鼠血脂水平,下调肝脏VCAM-1、ICAM-1基因表达,减轻肝损伤。AIM: To investigate the protective effect of Chunkangxuezhi capsule against liver injury in hyperlipemic rats. METHODS: Forty wistar male rats were randomly divided into 4 groups:blank group,model group,simvastatin group and Chunkangxuezhi capsule group. Hyperlipidemia model in rats was indeuced by hyperlipidemic diet for 3 weeks. Rats in each group were intragastric administrated with corresponding drugs. After 9weeks,the serum levels of total cholesterol( TC),triacylglycerol( TG),low-density lipoprotein cholesterol( LDL-C) and high-density lipoprotein cholesterol( HDL-C) were measured. Liver injury was evaluated with histopathologic examination by H. E.staining. The expression of intercellular adhesion molecule-1( ICAM-1) and vscular cell adhesion molecule-1( VCAM-1) in hepatic tissue were measured by immunohistochemical staining. RESULTS: Serum levels of TC,TG and LDL-C in the Chunkangxuezhi capsule group were significantly lower than those in the model group( all P 〈 0. 01),HDL-C was significantly lower than those in the model group( all P 〈0. 01),expressions of ICAM-1and VCAM-1 were significantly lower than those in the model group( all P 〈 0. 01). Liver injury in Chunkangxuezhi capsule group decreased than that in the model group.CONCLUSION: Chunkangxuezhi capsule can protect against liver injury via regulating expressions of ICAM-1and VCAM-1.
