摘  要:为了研究高原甘加藏羊发情周期垂体组织中促卵泡素β亚基(FSHβ)和促黄体素β亚基(LHβ)基因在m RNA水平的表达规律,选取发情周期不同阶段的高原甘加藏羊24只,从其垂体组织中提取总RNA,采用实时荧光定量PCR(q RT-PCR)技术,以β-actin为内参基因,比较研究了高原甘加藏羊发情周期垂体组织中FSHβ和LHβ基因在m RNA水平的表达差异。结果显示,FSHβ和LHβ的m RNA在高原甘加藏羊发情周期不同阶段的垂体组织中均有表达。发情前期、发情期、发情后期和间情期的FSHβm RNA相对表达量分别为0.235 9、1.0、3.547 5和3.666 7,发情前期与发情期、发情后期和间情期差异极显著(P<0.01),发情期与发情后期和间情期差异显著(P<0.05),发情后期与间情期差异不显著(P>0.05);LHβm RNA相对表达量为0.025 9、1.0、16.109 1和0.563 8,发情前期与发情期、发情后期和间情期差异极显著(P<0.01),发情期与发情后期差异极显著(P<0.01)、与间情期差异不显著(P>0.05),发情后期与间情期差异极显著(P<0.01)。因此,甘加藏羊发情周期内垂体组织中FSHβ和LHβ在m RNA水平上的表达呈现一种动态差异性变化趋势。本研究结果在分子水平上为揭示藏羊生殖内分泌调控机制提供了科学数据。The objective of this experiment is to examine the mRNA levels of FSH β gene and LH β gene in pituitary ofestrous cycle in Ganjia Tibetan sheep. The total RNA of pituitaries were sampled from 24 sheep in dif- ferent stages of estrous cycle and real-time fluorescence quantitative PCR (qRT-PCR) method with β-actin as reference gene was utilized to study above different expressiveness. The results showed that both FSH β gene and LH β gene were expressed in the pituitaries of Ganjia Tibetan sheep during the whole estrous cycle. The relative expression of FSH β mRNA in proestrus, estrus, metestrus and diestrus was 0.235 9,1.0,3.547 5 and 3.666 7, respectively, The expression of FSH β mRNA in proestrus was very significantly different from that other stage of estrous cycle (P〈0.01), and The expression of FSH β mRNA in oestrus was very significantly different from that in both metestrus and diestrus (P〈0.05),and The expression of FSH β mRNA in metestrus and diestrus was nosignificant difference (P〉 0.05 ).The relative expression of LH β mRNA in proestrus, estrus, metestrus and diestrus was 0.025 9,1.0,16.109 1 and 0.563 8, respectively,The LH β mRNA in proestrus was very significantly different from that other stage of estrous cycle (P〈0.01), The LH β mRNA the oestrus was significantly different in that of metestrus (P〈 0.01 ), but it was no significant difference in that of diestrus (P〉 0.05 ). The LH β mRNA was ex- tremely significant between metestrus and diestrus (P〈0.01).Therefore the relative expression of FSHfl gene and LH β gene in mRNA levels presented a dynamic variant trend in Ganjia Tibetan sheep.This experiments also pro- vided the scientific data in molecular levels to study reproductive endocrine regulation mechanism of Tibetan sheep.
