通心络对压力超负荷大鼠心肌HO-1mRNA表达及心功能的影响

摘  要:目的观察通心络对压力超负荷大鼠心肌组织中血红素加氧酶-1(HO-1)mRNA基因表达的影响,并探讨其对心功能的保护作用。方法选择40只SD大鼠,随机抽取8只作为假手术组,其余32只大鼠采用腹主动脉缩窄术制备压力超负荷模型,将术后存活的24只大鼠再随机分成模型组、通心络组及通心络+HO-1抑制剂组各8只。通心络组给予通心络超微粉水溶液1 g/(kg·d)灌胃,假手术组、模型组均给予等容积的生理盐水灌胃,通心络+HO-1抑制剂组给予通心络超微粉水溶液1 g/(kg·d)灌胃同时给予Zn PP注射剂1 mg/(kg·d)腹腔注射。各组每日9:00干预1次,连续干预4周后行心脏超声检测室间隔厚度(IVST)、左室后壁厚度(LVPWT)、左室舒张末内径(LVEDD)、左室收缩末内径(LVESD)和左室质量指数(LVMI)。8周后检测心肌组织HO-1mRNA表达水平,并在同时间段做心肌组织病理学HE染色。结果模型组、通心络组及通心络+HO-1抑制剂组IVST、LVPWT、LVEDD、LVESD及LVMI均显著高于假手术组(P均<0.05),通心络组上述各指标均明显低于模型组和通心络+HO-1抑制剂组(P均<0.05)。通心络组HO-1mRNA表达水平明显高于其他3组(P均<0.05);HE染色显示,通心络组和通心络+HO-1抑制剂组心肌组织形态学均较模型组有不同程度的改善,其中通心络组改善更为显著。结论通心络可以抑制心肌纤维化,减缓心室重构,保护心肌组织,此作用可能与通心络诱导HO-1过表达有关。Objective It is to observe the effect of Tongxinluo (TXL) on heme oxygenase- 1 (HO -1 ) mRNA gene ex- pression in myocardial tissue in the rats with pressure overload, and to investigate its protective effect on cardiac function. Methods 40 SD rats were selected, in which 8 ones were randomly selected as sham operation group, the other 32 rats were prepared abdominal aortic coarctation pressure overload models, the survival rats ( n = 24) after operation were randomly divid- ed into three groups: model group, TXL group and TXL + HO - 1 inhibitor group, each group had 8 rats. TXL group was giv- en TXL superfine aqueous 1 g/( kg · d) orally, sham operation group, model group were given the same volume of normal sa- line, TXL ·HO -1 inhibitor group was given Tongxinluo superfine aqueous 1 g/( kg ·d) orally + ZnPP injection 1 mg/( kg · d) by intraperitoneal injection. Each set of daily 9:00 medication once daily for 4 consecutive weeks, interventricular septum thickness (IVST) , left ventricular wall thickness ( LVPWT), left ventricular end diastolic diameter ( LVEDD), left ventricu- lar end systolic inner diameter (LVESD) and left ventricular mass index (LVMI) were detected by cardiac ultrasound. In 8 weeks the expression of HO - 1 mRNA in myocardial tissue was determined, and at the same time histopathological HE staining was used to observe pathologic change in myocardial tissue. Results Compared with sham operation group, IVST, LVPWT, LVEDD, LVESD and LVMI were significantly increased in model group,TXL group and TXL + HO - 1 inhibitor group, and these indexes in TXL group were significantly lower than that in model group and TXL + HO - 1 inhibitor group. HO - lmRNA expression level in TXL group was significantly higher than that in the other three groups. HE staining showed that compared with model group, myocardial tissue morphology were improved more or less in TXL group and TXL + HO - 1 inhibi- tor group, and the improvements in TXL group were the
