摘 要:目的了解本院临床分离肠球菌的生物膜形成能力以及其与毒力基因的相关性。方法收集温州医科大学附属第一医院2013年6月至2014年9月临床分离的肠球菌348株,其中粪肠球菌156株,屎肠球菌192株。采用结晶紫染色法测定肠球菌的生物膜形成能力,PCR方法筛查4种常见的肠球菌毒力基因esp、gel E、agg和ace的存在情况,统计分析肠球菌生物膜形成能力与毒力基因的关系。结果 348株肠球菌毒力基因esp、gel E、agg和ace的检出率分别为80.5%、46.0%、46.6%和44.3%,除esp基因外(P=0.078),粪肠球菌中的其他3种毒力基因的携带率均显著高于屎肠球菌(P<0.01);肠球菌的生物膜形成阳性率为54.6%(190/348),其中粪肠球菌的生物膜形成阳性率为75.6%(118/156),屎肠球菌的生物膜形成阳性率为37.5%(72/192);粪肠球菌esp、gel E和agg基因的携带与其生物膜形成阳性率密切相关,而屎肠球菌中,只有esp基因的携带与其生物膜形成阳性率密切相关(P<0.01)。结论本院分离的肠球菌具有较高的毒力基因携带率和生物膜形成能力,肠球菌毒力基因esp、gel E和agg的携带与其生物膜形成密切相关。Objective To investigate the ability of biofilm formation in Enterococcus isolated from our hospital and their correlation with virulence genes. Methods A total of 348 clinical Enterococcus isolates (156 Enterococcus faecalis and 192 Enterococcus faecium) were collected at the First Affiliated Hospital of Wenzhou Medical University in China during Jun 2013 to Sep 2014. The ability of biofilm formation assay were tested by crystal violet staining method, PCR method was used for screening of 4 common Enterococcus virulence factor genes, and then the relationship between the ability of biofilm formation and virulence factor genes was analysed. Results The rates of virulence factor genes (esp, gelE, agg and ace) of 348 strains of Enterococcus were 80.5%, 46.6%, 46% and 44.3% respectively. Except for the esp gene (P=0.078), the rate of other 3 virulence factor genes in Enterococcus faecalis were significantly higher than that in Enterococcusfaecium (P〈0.01). The positive rate of biofilm formation of Enterococcus was 54.6% (190/348), whereas the positive rate of biofilm formation in Enterococcusfaecalis was 75.6% (118/156), and the positive rate of biofilm formation in Enterococcus faecium was 37.5%(72/192). Enterococcus faeealis virulence factor genes esp, gelE and agg were all closely related to biofilm formation, and Enteroeoeeusfaecium virulence factor gene esp was closely related to biofilm formation (P〈0.01). Conclusion The carrying rate of virulence genes and the ability of biofilm formation were high in Enterococcus isolated in our hospital. Enterocoeeus virulence factor genes esp, gelE and agg were all closely related to biofilm formation.
