摘 要:目的基于NF-κB信号通路观察小青龙汤联合玉屏风散对变应性鼻炎(allergic rhinitis,AR)大鼠鼻黏膜AQP5、NF-κB p65和p-CREB表达的影响,揭示小青龙汤联合玉屏风散治疗AR的作用机制及以饮论治AR的合理性。方法将清洁级健康雄性Wistar大鼠48只随机分为6组,即分为正常组、AR模型组、Forskolin干预组、H89干预组、PDTC干预组、小青龙汤联合玉屏风散组,每组8只。采用卵蛋白全身致敏与局部攻击方法制作AR大鼠模型,观察小青龙汤联合玉屏风散组在治疗后鼻黏膜AQP5、NF-κB p65和p-CREB的表达。结果模型组AO、P5、p-CREB表达下调;Forskolin干预组、PDTC干预组、小青龙汤联合玉屏风散组均能使AO,P5、p-CREB表达上调,而cAMP依赖蛋白激酶(protein kinase A,PKA)抑制剂H89能下调AQP5、p-CREB表达。模型组NF-κB p65表达上调,Forskolin干预组、PDTC干预组、小青龙汤联合玉屏风散组均能使NF-κB p65表达下调,而PKA抑制剂H89能上调NF-κB p65表达。结论小青龙汤联合玉屏风散治疗变应性鼻炎作用机制可能与抑制NF-κB信号通路和兴奋cAMP-PKA信号通路使AQP5表达上调有关,为以饮论治AR奠定了现代生物学基础。Objective To observe the effect of the Xiaoqirtglong decoction and Yupirt^eng power on the expression of AQP5 ,NF- kappaB p65 and p-CREB in nasal mucosa of allergic rhinitis rats in NF- kappaB signaling pathway, and reveal the mechanism of the Xiaoqinglong decoction and Yupingfeng power in treating AR and the rationality of treating AR by phlegm and retained fluid. Methods 48 healthy male Wistar rats were divided into 6 groups by randomized boeks design: normal group, AR model group, Forskolin ( cAMP activator) group, H89 ( PKA inhibitor) group, PDTC ( NF-kappa B inhibitor) , and the Xiaoqinglong decoction and Yupingfeng power group, 8 rats in each group. The AR rat model was made by the method of whole body sensitization and local attack. The expression of AQPS, NF- kappaB p65 and p- CREB were observed after the treatment. The results were statistically analyzed. Results The expression of AQP5 and p-CREB in the model group was decreased, and the expression of AQP5 and p-CREB in the Forskolin group, PDTC group, the Xiaoqinglong decoction and Yupingfeng power group was increased, while the PKA inhibitor H89 could decrease the expression of AQPS. The expression of NF- kappaB p65 in the model group was increased, and the expression of NF- kappaB p65 in the Forskolin group, PDTC group, the Xiaoqinglong decoction and Yupingfeng power group was decreased, while the PICA inhibitor H89 could increase the expression of NF- kappaB p65. Conclusion Xiaoqinglong decoction and Yupingfeng powder treatment of allergic rhinitis mechanism may be related to the inhibition of NF- kappa B signaling pathway and cAMP-PKA signaling pathway to increase the AQP5 expression, which has laid the foundation of modern biology for treating AR by phlegm and retained fluid.
