实时荧光定量PCR模板核酸提取方法的比较

摘　　要：对病料核酸提取前处理方法进行优化,选择酚/氯仿法、试剂盒提取法和DNA/RNA同提取法,比较提取效果,为实验室核酸提取提供参考。配制模拟样品,通过实时荧光定量PCR比较3种方法提取效果。结果表明,提取纯度为试剂盒提取法>酚/氯仿法>DNA/RNA同提取法。提取效率为酚/氯仿法>DNA/RNA同提取法>试剂盒提取法。表明各提取方法效果差异显著(P<0.01),为实验室核酸提取提供了参考。To obtain the appropriate nucleic acid sample,the pretreatment method for extracting nucleic acid from the disease-tissue is optimized,the extraction efficiencies of phenol/chloroform method,reagent box extraction method and extraction method with DNA and RNA were compared by simulated samples and Green I SYBR real-time PCR.The results showed that the extraction purity（Reagent box extraction method〉Improved phenol/chloroform method〉Extraction method with DNA and RNA）and the extraction efficiency（Improved phenol/chloroform method〉Extraction method with DNA and RNA 〉Reagent box extraction method）were different.The experiment showed that the extraction methods had a significant difference,which provided a reference for laboratory nucleic acid extraction.
