摘 要:目的 观察人降钙素基因相关肽α(hCGRPα)转染骨髓间充质干细胞(BMSCs)联合自体骨移植在兔股骨头坏死(ONFH)模型中成骨基因的表达.方法 采用液氮冷冻法成功制作出家兔股骨头坏死兔.将72只ONFH模型兔随机分为3组.hCGRPα基因转染BMSCs组(A组):将hCGRPα基因转染BMSCs移植于股骨头内;BMSCs组(B组):植入BMSCs;无关序列组(C组):植入干细胞为无关基因BMSCs.3组中均同时将自体骨植入股骨头.于实验第1个月末观察兔股骨头的细胞内hCGRPα和成骨基因骨钙素的表达.结果 将hCGRPα基因转染BMSCs植入股骨头1个月时,A、B、C组中hCGRPα mRNA与其蛋白的相对表达量分别为0.84±0.06、0.31±0.02、0.29±0.02与0.76±0.07、0.21±0.02、0.18±0.02.3组中骨钙素mRNA与其蛋白的相对表达量分别为0.76±0.07、0.39±0.04、0.29±0.04与0.68±0.06、0.33±0.03、0.31±0.03.A组股骨头细胞中hCGRPα基因和骨钙素基因均呈高表达,明显高于B组和C组,差异均有统计学意义(P<0.05).结论 hCGRPα修饰BMSCs植入ONFH动物模型股骨头中,可促进细胞中成骨因子hCGRPα和骨钙素基因的表达.Objective To observe the expression of osteogenic gene in the femoral head after transplantation of calcitonin gene-related peptide α (hCGRPαt)-transfected bone marrow mesenchymal stem cells (BMSCs) combined with bone graft into osteonecrosis of the femoral head (ONFH) model of rabbits.Methods The osteonecrosis models on one side femoral head of rabbits were established by liquid nitrogen.Seventy-two rabbit ONFH models were randomly divided into 3 groups.In group A (hCGRPα-transfected BMSCs group),the hCGRPα was transfected into BMSCs.In group B (BMSCs group),BMSCs were implanted.In group C (irrelated sequence group),BMSCs with irrelated gene (only pLNCX2-GFP) were implanted.In 3 groups,autologous bone grafts were implanted into the femoral heads simultaneously.After one month,the expression of hCGRPα and osteocalcin mRNA and proteins in the cells of rabbit femoral head was observed respectively.Results At 1 st month,the expression levels of hCGRPα mRNA and protein were 0.84 ±0.06,0.31 ±0.02,0.29 ±0.02 and 0.76 ±0.07,0.21 ±0.02,0.18 ±0.02 respectively in groups A,B,and C.The expression levels of osteocalcin mRNA and protein were 0.76 ± 0.07,0.39 ± 0.04,0.29 ± 0.04 and 0.68 ± 0.06,0.33 ± 0.03,0.31 ± 0.03 respectively in groups A,B,and C.The expression levels of hCGRPα and osteocalcin mRNA and proteins in group A showed highest among the three groups (P 〈 0.05).Conclusion The transplantation of hCGRPα-transfected BMSCs combined with bone graft into ONFH animal model can promote the expression of osteogenic factor hCGRPα and osteocalcin gene in cells of the femoral head.
