HPLC法同时测定藏药独一味中山栀苷甲酯、木犀草素、芹菜素的含量

摘　　要：目的：建立同时测定独一味中山栀苷甲酯、木犀草素、芹菜素含量的方法。方法：采用高效液相色谱法。色谱柱为Diamonds C18,流动相为乙腈-0.1%磷酸溶液（梯度洗脱）,流速为1.0 ml/min,检测波长为238 nm（山栀苷甲酯）、348 nm（木犀草素、芹菜素）,柱温为25℃,进样量为10μl。结果：山栀苷甲酯、木犀草素、芹菜素检测质量浓度线性范围分别3.08-308μg/ml（r=0.999 8）、3.72-372μg/ml（r=0.999 9）、2.92-292μg/ml（r=0.999 9）;精密度、稳定性、重复性试验的RSD〈3.0%;加样回收率分别为95.38%-103.15%（RSD=1.85%,n=9）、96.05%-102.66%（RSD=2.07%,n=9）、95.49%-103.59%（RSD=2.18%,n=9）。结论：该方法操作简便,精密度、稳定性、重复性好,可用于独一味中山栀苷甲酯、木犀草素、芹菜素含量的同时测定。OBJECTIVE： To establish a method for the contents determination of dipsacoside B, luteolin and apigenin in Zang medicine Lamiophlomis rotata. METHODS： HPLC was performed on the column of Diamonds C18 with mobile phase of acetonitrile-0.1% phosphoric acid solution （gradient elution） at a flow rate of 1.0 ml/min, detection wavelength was 238 nm for dipsacoside B, 348 nm for luteolin and apigenin, column temperature was 25 ℃, and injection volume was 10μl. RESULTS： The linear range was 3.08-308 μg/ml for dipsacoside B （r=0.999 8）, 3.72-372μg/ml for luteolin （r=0.999 9）and 2.92-292 μg/ml for apigenin（r=0.999 9） ; RSDs of precision, stability and reproducibility tests were lower than 3.0% ; recoveries were 95.38%-103.15% （RSD= 1.85%, n=9）, 96.05% -102.66% （RSD=2.07%, n=9） and 95.49% -103.59% （RSD=2.18%, n=9）. CONCLUSIONS： The method is simple with good precision, stability and reproducibility, and can be used for the simultaneous determination of dip- sacoside B, luteolin and apigenin in Zang medicine L. rotata.
