摘  要:猪塞内加谷病毒(Seneca Valley virus,SVV)是一种新出现的可感染仔猪及母猪并导致仔猪死亡的病毒。2015年3月份至今年在我国、巴西及美国出现了几起塞内加谷病毒感染猪群并伴随严重的临床症状及发病死亡的疫情,造成严重的经济损失。为分离鉴定SVV,本研究将RT-PCR检测为SVV阳性的猪临床样品无菌处理后接种PK-15细胞,连续传代培养。通过细胞病变、RT-PCR扩增、电镜观察和基因序列测定对细胞培养物进行鉴定。结果表明我们成功分离到国内首株SVV,并将其命名为SVV CH-01-2015。全基因组的系统发育分析表明SVV CH-01-2015位于Senecavirus病毒属,与Senecavirus病毒属中的病毒成员同源性最高,由此可以确定本研究所分离到的SVV属于Senecavirus病毒属。本研究为进一步研究SVV的致病性和致病机制奠定了基础。Seneca Valley virus (SW) is a novel emerging virus, which was able to infect piglets and sow, but only resulting in the death of the piglets. Since March 2015 to this year, several outbreaks of SVV infections in pig herds have occurred in China, Brazil and United States, respectively, causing heavy losses. To isolate and identify the virus in China, porcine kidney cell (PK-15) monolayers were inoculated with the clinical samples that were positive for SVV detected by RT-PCR. Subsequently, 4 isolates of SVV were isolated, which induced cytopathic effect at the first passage on PK-15 cells and were further confirmed by RT-PCR, electron microscopy examination and gene sequencing, designated CH-01-2015, CH-02-2015, CH-03-2015 and CH-04-2015, respectively. Phylogenetic analysis based on whole genome indicated that the SVV isolates belonged to a member of the Senecavirus. The results of this study are critical for the further studies of pathogenicity and pathogenic mechanisms of SVV.
