摘 要:为分析死亡食蟹猴脑组织菌群结构情况,初步探索细菌对食蟹猴的致死因素,本研究按照标准化操作流程收集死亡食蟹猴的大脑、小脑病理组织样本,提取样本中全部细菌总DNA,利用通用引物27F/338R PCR扩增细菌16S rDNA V1-V2高变区。扩增产物经高通量测序,并利用QIIME软件对测序数据进行生物信息学分析。结果显示,测序共生成238 Mbp原始数据,获得脑组织细菌的结构组成,其中优势菌门包括厚壁菌门、拟杆菌门和变形菌门,优势菌科包括普雷沃氏菌科、瘤胃菌科、鞘氨醇单胞菌科等,含量最高的细菌属为普雷沃菌属、螺旋体属、交替假单胞菌属等。本研究结果表明死亡食蟹猴的脑组织中含有多种细菌,可能会引起感染和炎症,结合病理学检查,提示食蟹猴脑组织病原菌感染可能与死亡的发生存在一定关联。This study aimed to analyze the flora structure in brain tissue of a dead cynomolgus monkey and to explore the lethal factors, so as to develop for potential prevention and treatment of the diseases. The pathological cerebrum and cerebellum tissue samples were collected according to the standardization process. Total bacterial DNA were extracted and universal primers 27F/338R were chosen to amplify the V1 and V2 hypervariable region of bacterial 16S rDNA in samples by PCR technique. The PCR products were sequenced by high-throughput sequencing (HTS). HTS generated 238 Mbp raw sequencing data, which were analyze by QIIME software to generate the microbial profiles from the infected cerebrum and cerebellum tissues. The results desplayed the microbial profiles in phylum, family, genus levels, respectively. The dominate phylums were Firmicutes, Bacteroidetes and Proteobacteria, and the dominate family were Prevotellaceae, the dominate genus were Prevotella, Treponema and and Sphingomonadaceae, and The sequencing results showed that the dead monkey was probably caused by infection of several bacteria, which induced inflammation. By combining with pathological examination, the results may shield light on the relation between the bacterial infection and death.
