摘  要:目的:探讨欧亚旋覆花总黄酮(Inula britannica flower total flavonoids,IBFTF)对D-半乳糖诱导的亚急性衰老模型小鼠的抗衰老作用。方法:随机将昆明小鼠分为6组(每组10只):空白对照组,模型组,维生素E(vitamin E,VE)组,欧亚旋覆花总黄酮低、中、高剂量组。空白对照组给予颈背部皮下注射0.3 m L生理盐水,其余组均给予等体积500 mg/kg D-半乳糖,每日1次,连续8周。造模第3周,低、中、高剂量组分别灌服0.5 m L欧亚旋覆花总黄酮100、200和400 mg/kg,VE组灌服等体积维生素E 100 mg/kg,空白对照组和模型组灌服等体积生理盐水,每日1次,连续6周。检测小鼠体质量、胸腺指数和脾指数,同时测定血清中抗氧化酶活性如超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)、过氧化氢酶(catalase,CAT)和谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)等衰老指标。β半乳糖苷酶染色试剂盒检测小鼠海马组织中衰老相关的β半乳糖苷酶含量。结果:低、中、高剂量组小鼠体质量明显高于模型组,差异具有统计学意义(P=0.053、P=0.001、P=0.000);低、中、高剂量组小鼠胸腺指数和脾指数明显高于模型组(均P=0.000);低、中、高剂量组小鼠血清中SOD、CAT和GSHPx活力明显高于模型组,其MDA含量低于模型组(均P=0.000);低、中、高剂量组小鼠海马组织β半乳糖苷酶(senescence-associated beta-galactosidase,SA-β-gal)阳性率明显低于模型组,差异有统计学意义(P=0.001、P=0.000、P=0.000)。结论:欧亚旋覆花总黄酮是一种天然抗氧化剂,可延缓D-半乳糖诱导的模型小鼠的机体衰老。Objective:To investigate the anti-aging effect of Inula britannica flower total flavonoids(IBFTF)on mice in D-galactose induced aging model. Methods:Kunming mice were randomly divided into 6 groups(10 mice in each group):control group,model group,vitamin E(VE) group,low,medium and high dose of IBFTF group. Mice in control group were subcutaneously injected with 0.3 m L saline,while those in other groups were injected with the same volume of D-galactose(500 mg/kg),once daily for 8 weeks. From the third week,mice in low,mediumand high dose group were orally administered with 0.5 m L 100,200,400 mg/kg IBFTF;those in VE group received 100 mg/kg vitamin E via intragastric administration;those in control group and model group were orally administered equal volume of saline once a day for 6 weeks. Body weight,thymus index and spleen index were detected,and antioxidant enzymes activities in serum such as superoxide dismutase(SOD),malondialdehyde(MDA),catalase(CAT) and glutathione peroxidase(GSH-Px) were also measured. Senescence-associated beta- galactosidase( SA- β- gal) activity in hippocampus was determined using a SA- β- gal staining kit. Results:The body weight of mice in low,medium and high dose IBFTF groups were significantly higher than that in the model group,with statistically significant difference(P=0.053,P=0.001,P=0.000). The thymus indexes and spleen indexes of mice in low,medium and high dose IBFTF groups were significantly higher than those in model group(P=0.000). The activity of GSH-Px,CAT and SOD in the serum of low,medium and high dose IBFTF groups was significantly higher than that in the model group,while the MDA content was lower than that in the model group(P=0.000);β-galactosidase positive rate in hippocampus of low,medium and high dose IBFTF groups was significantly lower than that in model group,with statistically significant differences(P=0.001,P=0.000,P=0.000).
