摘 要:目的采用PCR-DGGE法探讨纳米山药多糖靶向制剂对肠道微生态失调模型大鼠的调节作用。方法以i.g盐酸林可霉素造成大鼠结肠炎模型,将大鼠随机分成常规治疗组,靶向制剂组和自然恢复组,分别给予相应药物治疗。灌胃10d后处死所有大鼠,采集盲肠内容物,提取总DNA,PCR-DGGE方法检测样本菌群DNA。将所有数据收集得到DGGE凝胶,采用Quantity One软件进行相似性和多样性分析,观察纳米山药多糖靶向制剂对肠道微生态失调大鼠的治疗效果。结果样本提取总DNA浓度值(ng/μL)分别为:靶向制剂组(0.180±0.005)和常规治疗组(0.175±0.006)高于自然恢复组(0.072±0.001)(P<0.05),与正常水平(0.182±0.006)相近;DGGE的多样性指数分析结果为:靶向组(23.95±2.36)和常规治疗组(24.00±1.68)的条带丰富度大于自然恢复组(19.57±2.52),差异有统计学意义(P<0.05)且接近正常对照组(25.87±2.10)。结论纳米山药多糖靶向制剂组对肠道微生态失调大鼠具有显著的调整作用,是理想的中药微生态调节剂。Objective To explore the regulatory effects of nano Chinese yam polysaccharides(CYPs)synbiotics on dysbacteria in rats by PCR-DGGE.Methods The rat models of dysbacteria were established by intragastric administration of lincomycin,and randomly divided into the conventional treatment group,nano CYP group,or spontaneous recovery group to accept corresponding treatment respectively.All the rats were sacrificed after ten days of gavage;the cecal contents were collected to extract total DNA,and PCR-DGGE method was used for detection of the bacterial DNA.Finally,the Quantity One software was used to analyze the similarity and diversity,and the effects of nano CYPs on dysbacteria of rats were observed.Results The concentration of total DNA(ng/μL)was(0.180±0.005)in the nano CYP group,and(0.175±0.006)in conventional treatment group,which were higher than the(0.072±0.001)in spontaneous recovery group(P〈0.05),and similar to the(0.182±0.006)in normal control group.DGGE analysis showed that the diversity index of the nano CYP group(23.95±2.36)and that of the conventional treatment group(24.00±1.68)were greater than that of the spontaneous recovery group(19.57±2.52)with significant difference(P〈0.05),while similar to that of the normal control group(25.87±2.10).Conclusion The colon targeting nano CYPs,as an ideal microecological modulator,can significantly modulate intestinal microecological imbalance in Wistar rats.
