摘 要:以西瓜皮为原料,探讨其培养液对草履虫种群生长的影响。将新鲜西瓜皮配成2%、4%、6%、8%、10%、12%、14%、16%、18%、20%的培养液,进行草履虫培养试验,在接种后第4、7、10、16、19、30天采用微量计数法对草履虫进行计数,培养期为30 d。试验结果表明:2%、4%、6%、8%的西瓜皮培养液中草履虫达到最大密度值一般在接种后的10 d,其最大种群密度分别为(533.36±26.47)、(666.70±15.46)、(920.10±16.47)、(880.57±46.35)m L^(-1);而10%、12%、14%、16%的西瓜皮培养液达到最大密度一般在接种后7 d,其最大种群密度分别为(640.53±24.21)、(741.32±28.32)、(862.07±35.19)、(912.86±35.86)m L^(-1)。16 d时,所有培养液中草履虫密度开始急速下降;到19 d时,10%、12%、14%、16%的培养液中已经检测不到草履虫了;到30 d时,只有在2%、4%、6%、8%中检测到少量草履虫。从整个培养期来看,6%和8%的西瓜皮培养液培养草履虫效果最佳,最佳培养时间为7~10 d。本试验旨在为生物试验教学及科学研究中培养草履虫提供快速、经济、可行的培养方法。Ten kinds of mediums were used in the study, they were respectively 2%, 4%, 6%, 8% , 10%, 12% , 14% , 16%, 18% , 20% media of watermelon rind, being used to culture paramecium (Paramecium caudatum) , to compare the effects of different mediums of watermelon rind on the population growth of paramecium and select the suitable mediums for culturing parameci- um. After culturing paramecium of 4 d, 7 d, 10 d, 16 d, 19 d, 30 d, micro-counting method- were used and cultivation period was 30 days. The results showed that 2% , 4% , 6% , 8% of watermelon rind mediums, paramecium maximum density were respectively (533.36±26.47 ), (666.70± 15.46), (920.10 ± 16.47) and (880. 57±46.35) mL^-1 at 10 days. The maximum density of concentration of 10%, 12% , 14% , 16% watermelon rind mediums were respectively (640.53 ±24.21), (741.32±28.32), (862.07±35.19) and (912.86±35.86) mL^-1 at7 days. At 16 days, paramecium density in all cultures began to fell sharply at 10% , 12% , 14%,12% of medium. At 30 day, 2% , 4% , 6% , 8% of watermelon rind mediums could detect a few paramecia. From whole period, the most suitable concentration of watermelon rind mediums were 6% and 8% and the best time was 7 to 10 days. This experiment was intended to cultivate paramecium in biological experiment teaching and scientific research to provide fast, economic and feasible methods for culturing paramecium.
