摘 要:观察白屈菜红碱预处理对糖尿病大鼠肾缺血再灌注心肌损伤的影响,并探讨其可能的作用机制.成年SD大鼠分为正常组、糖尿病组、缺血再灌注组和白屈菜红碱预处理+缺血再灌注组,各组动物经1%戊巴比妥钠腹腔麻醉,开腹,暴露双侧肾动、静脉,各组经缺血再灌注或经白屈菜红碱预处理后检测相应试验指标.结果显示糖尿病大鼠肾脏缺血再灌注后,I/R组肾功能明显下降,BUN和Scr均明显高于sham和DM组,心肌组织SOD活力较sham和DM组明显降低,LDH、CK活力和MDA含量显著升高.与I/R组相比,白屈菜红碱组BUN、Scr显著减少,SOD活力明显增加,LDH、CK活力和MDA含量明显降低.白屈菜红碱组心肌组织NF-κB表达降低.得出结论,缺血再灌注后,肾功能和心功能明显受损.白屈菜红碱预处理可以减轻糖尿病大鼠肾缺血再灌注远程心肌损伤,改善心肌功能.This papers aims to observe the effects with Chelerythrine pretreatment on renal ischemia/ reperfusion myocardium injury in diabetic rats and explore its possible mechanism. Adult SD rats are divided into normal group, diabetes mellitus group, ischemia reperfusion group and Chelerythrine pretreatment+ ischemia reperfusion group. Rats in each group are opened and their bilateral renal arteries and veins are exposed after in- traperitoneal anesthesia by 1% of Pelltobarbitalum Natricum and the relevant experiment indexes are tested in each group after ischemia/reperfusion or/and Chelerythrine pretreatment. The results show that after renal ischemia/reperfusion in diabetic rats, renal function of I/R group decreases in a significant way, BUN and Scr are much higher than those of sham and DM groups; the activity of SOD in myocardial tissue decreases to a greater extent than that of sham and DM group; the activity of LDH and CK and the content of MDA increases noticeably. Comparing with I/R group, BUN and Scr in chelerythrine+DM+ I/R group significantly reduces, but the activity of SOD increases obviously, and the activity of LDH and CK and the content of MDA decrease to a greater degree; and the expression of NF-kB in the myocardium decreases. In conclusion, after ischemia/ reperfusion, renal function and cardiac function are impaired; Chelerythrine pretreatment can reduce myocardial injury after renal ischemia reperfusion which causes distance injury in diabetic rats, and improve myocardial function.
