加味宫外孕Ⅱ号方对早孕大鼠滋养细胞及肝脏的影响

摘　　要：目的观察加味宫外孕Ⅱ号方对早孕大鼠滋养细胞及肝脏的影响。方法将早孕大鼠分为西药组(甲氨蝶呤)、中药(加味宫外孕Ⅱ号方)低、中、高剂量组、中西药结合组和空白组(生理盐水),分别予甲氨蝶呤肌注、中药或生理盐水灌胃,第8天处死孕鼠取血、肝脏、子宫及孕囊,检测血β-HCG,观察滋养细胞及肝脏组织病理学改变。结果与空白组比较,另5组出现滋养细胞结构异常、破坏,线粒体肿胀,核异染色质增多,炎细胞浸润及血β-HCG下降,以中药高剂量组和中西药结合组较明显,与其他3组比较差异有统计学意义(P<0.05),中药中剂量与低剂量组比较差异有统计学意义。除中药低剂量组外,另外几组孕鼠肝脏病理学评分上升,与空白组比较差异有统计学意义,西药组和中西药结合组上升明显,与中药高、中剂量组比较差异有统计学意义。结论加味宫外孕Ⅱ号方通过影响孕鼠滋养细胞形态结构及功能而达到损坏胚胎目的,且存在量效关系,剂量增高作用增强;而对孕鼠肝损害较甲氨蝶呤轻。Objective To observe the influence of trophoblast cell and liver of pregnancy rats with the modified decoction Ⅱfor ectopic pregnancy.Methods The building successful pregnancy rats were randomly divided into the western medicine group（treat by methotrexate）,the Chinese medicine group（The modified decoctionⅡfor ectopic pregnancy low,medium,high dose）,the combination of Chinese and western medicine group（treat by methotrexate and Party）,and the blank group（saline）.These groups were given an intramuscular injection with MTX,Party or saline was gavage once a day,for 7days.On the eighth day of drug delivery,the rats were killed and taken the femoral arterial blood,livers,gravid uterus and gestational sacs,and measured the value of bloodβ-HCG,then observed the pathology change of trophoblast cells and liver tissue.Results Compared with the blank group,the other five treatment groups of trophoblast cells structure were damaged to varying degrees,mitochondria swelling,nucleus heterochromatin increased,inflammatory cells infiltration,the structure of embryo abnormalities and reduced bloodβ-HCG value.The changes in TCM high dose group and the combination of Chinese and western medicine group were more apparent.There were differences in comparison with the other three treatment groups（P〈0.05）.And there were differences in comparison with the TCM medium dose group and the low dose group.In addition to TCM low dose group,the other groups of pregnant rats liver pathology score rise,there were statistical significantly differences compared with the blank group.The score of western medicine group and the combination of Chinese and western medicine group increased significantly,which were differences compared with the TCM high and medium dose group.Conclusion Modified decoction Ⅱfor ectopic pregnancy destructed trophoblast morphological structure,reducing the amount of sertoli cell,normal physiological function damaged and achieve the goal of killing embryos.There is concentration-response relationsh
