美洲大蠊提取物对乙醇致小鼠急性胃溃疡的预防作用

摘　　要：目的考察美洲大蠊提取物及其分段分子量(M)部分对乙醇致小鼠急性胃溃疡的预防作用,并探讨其作用机制。方法将80只SPF级ICR雄性小鼠随机分为空白组,模型组,康复新液组,美洲大蠊提取物组以及M<1 000 Da、1 000 Da<M<5 000 Da、5 000 Da<M<10 000 Da、M>10 000 Da部分组,每组灌胃给予相应药物,连续给药7 d。除空白组外,其余组无水乙醇造模,处死动物后测定溃疡抑制率,进行胃黏膜病理学观察,测定相应指标含有量或活性。结果与空白组相比,模型组小鼠胃黏膜溃疡病变明显,炎性细胞浸润明显增多,脂质过氧化物丙二醛(MDA)、一氧化氮(NO)及诱导型一氧化氮合酶(iNOS)含有量明显升高,而组成型一氧化氮合酶(c NOS)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)及谷胱甘肽过氧化物酶(GSH-Px)活性明显下降。与模型组相比,美洲大蠊提取物及其1 000 Da<M<5 000 Da部分溃疡抑制作用明显,改善了小鼠胃组织的病理学状态,还可显著降低胃黏膜损伤组织内MDA、NO和iNOS水平,提高cNOS、SOD、CAT和GSH-Px活性。结论美洲大蠊提取物可通过降低氧自由基及NO自由基的含有量而预防急性胃溃疡,其中1 000 Da<M<5 000 Da部分作用最为明显。AIM To investigate the protective effects of Periplaneta americana L. extract and its segment molecular weight（ M） parts on mice with ethanol-induced acute gastric ulcer and its mechanism of action. METHODS Eighty SPF ICR male mice were randomly divided into control,model,Kangfuxin Oral Liquid,P. americana extract,M 〈 1 000 Da,1 000 Da 〈 M 〈 5 000 Da,5 000 Da 〈 M 〈 10 000 Da and M 〉 10 000 Da parts groups. Intragastric administration of corresponding drugs were given to each group for successive seven days. Except for the control group,the models in another groups were established by anhydrous ethanol. Then all the animals were sacrificed,ulcer inhibition rate was detected,pathology observation of gastric mucosa was carried out,and contents or activities of relevant indices were determined. RESULTS Compared with the control group,the gastric mucosa of mice in model group showed obvious ulcer lesions,and infiltration of inflammatory cells was increased obviously,contents of lipid peroxides including malondialdehyde（ MDA）,nitrc oxide（ NO） and inducible nitric oxide synthase（ iNOS） were increased significantly,while activities of constitutive nitric oxide synthase（ cNOS）,superoxide dismutase（ SOD）,catalase（ CAT） and glutathione peroxidase（ GSH-Px） were decreasedsignificantly. Compared with the model group,the inhibitory effects of P. americana extract and its 1 000 Da 〈M 〈 5 000 Da part on ulcer were obvious,the pathological state of mice’s stomach tissue was improved,the levels of MDA,NO and iN OS were obviously decreased,and the activities of c NOS,SOD,CAT and GSH-Px were significantly increased. CONCLUSION P. americana extract can prevent acute gastric ulcer by decreasing the contents of oxygen free radicals and NO free radical oxidations,especially 1 000 Da 〈 M 〈 5 000 Da part.
