摘  要:目的利用代谢组学方法,评价扶正化瘀方(丹参、桃仁、松花粉等)对大鼠肝纤维化的治疗效果,并探索其可能的机制。方法将SD大鼠随机分成正常对照组(n=6)、二甲基亚硝胺(DMN)诱导肝纤维化模型组(n=6)及扶正化瘀方给药组(n=6),采用超高效液相色谱-飞行时间质谱(UHPLC-TOF-MS)技术对大鼠血清进行分析,所得数据采用偏最小二乘判别分析法(PLS-DA)进行模式识别处理。结果正常组和模型组大鼠的血清代谢轮廓存在显著差异,而给药组在空间投影上呈现出"偏离模型组且接近正常组"的趋势。根据变量重要性投影值(VIP),筛选出19种相关代谢标志物。结论验证了扶正化瘀方对大鼠肝纤维化的治疗效果,其潜在机制可能与修复体内异常的脂质和氨基酸代谢有关。AIM To evaluate the therapeutic effect of Fuzheng Huayu Formula( Salviae miltiorrhizae Radix et Rhizoma,Persicae Semen,Pini Pollen,etc.) on rats with liver fibrosis by metabonomics and to investigate its possible mechanism. METHODS SD rats were randomly divided into control group( n = 6),liver fibrotic model group induced by dimethylnitrosamine( DMN)( n = 6) and Fuzheng Huayu Formula intervention group( n =6). The rat sera were analyzed by ultra-high performance liquid chromatography-time-of-flight-mass spectrometry( UHPLC-TOF-MS),and the obtained data were processed by partial least squares-discriminant analysis( PLSDA) to identify patterns. RESULTS The significant differences of serum metabolic profiles were observed between the control and model groups. Fuzheng Huayu Formula intervention group exhibited a tendency to move away from the model group and near the control group in space projection. Nineteen relevant metabolic makers were then screened out according to their variable importance in the projection( VIP) values. CONCLUSION Therapeutic effect of Fuzheng Huayu Formula on liver fibrosis in rats is demonstrated,and its potential mechanism may be associated with restoring abnormal lipid and amino acid metabolisms.
