摘 要:目的探讨丙烯腈(acrylonitrile,ACN)对大鼠的神经行为及脑组织生化指标的影响,为ACN神经毒性的研究提供线索。方法将40只SPF级SD大鼠按体重随机分为4组:3个ACN染毒组(12.5、25、50 mg/kg)、对照组(玉米油)灌胃染毒,1次/d,6天/周,连续13周。末次染毒后,使用Video Track动物行为分析系统对其自主活动度进行检测,同时检测大鼠脑组织中乙酰胆碱酯酶(ACh E)活力、乳酸和葡萄糖含量及血糖含量。结果染毒6周后,高剂量组出现明显流涎。各组之间脑组织脏器系数比较,差异均无统计学意义(P>0.05)。大鼠脑组织中ACh E活力低、中、高剂量组与对照组相比降低(P<0.05);乳酸含量中、高剂量组与对照组和低剂量组相比降低(P<0.05);葡萄糖含量各组间比较,差异均无统计学意义(P>0.05)。大鼠血糖含量高剂量组明显低于对照组、低和中剂量组(P<0.05)。平面翻正和断崖回避试验中,染毒前和染毒后各组大鼠结果均为阳性。Openfield试验中各组大鼠在每个时间段的运动距离比较,差异均无统计学意义(P>0.05)。结论在该试验条件下ACN能够引起脑组织ACh E活力、乳酸含量及血糖含量的变化,但未引起大鼠自主活动功能和活动量的变化。[Objective]To explore the effects of acrylonitrile(ACN)on nervous behavior and brain biochemical indexes in rats,provide a clue for the research of neurotoxicity of ACN. [Methods]40 SD rats(SPF degree)were randomly divided into four groups by weight,including 3ACN groups(12.5 mg/kg,25 mg/kg,50 mg/kg) and a control group(corn oil). The animals were treated by intragastric administration once a day,6 days a week for 13 weeks. After the last gavage,the autonomous activity of rats was measured by Video Track Automated Behavioral Analysis Systems. ACh E activity,lactic acid content and glucose content in brain,and blood sugar content were detected.[Results]After six weeks of exposure,the obvious salivation phenomenon was observed in the high-dose group. The difference in brain organ coefficient was not statistically significant among different groups(P〈0.05).ACh E activity in three ACN groups was lower than that in the control group(P〈0.05).The lactic acid content in the middle-dose group and high-dose group was lower than that in the low-dose group and the control group(P〈0.05).There was no statistically significant difference in glucose content among different groups(P〈0.05).The blood sugar level in high-dose group was significantly lower than that in the low-dose group,middle-dose group and the control group(P〈0.05).The results of the surface righting reflex test and cliff avoidance reflex test were positive in all groups before and after exposure. There were no statistically significant differences in motion distance of every minute in Openfield test among all groups(P〈0.05).[Conclusion]Under the test conditions,ACN can lead to the change of ACh E activity and lactic acid content in brain,as well as blood sugar level,but there is no obvious effect on the autonomic activity and activity quantity.
