Effect of Taohong Siwu Decoction(桃红四物汤) on Angiogenesis and MMP-2 in Rabbit Skin Graft Model

摘  要:目的:观察桃红四物汤对兔自体皮片移植术后移植皮片的血管密度、血清中MMP-2含量的影响,探讨桃红四物汤对皮片移植术后血管新生的影响机制。方法:将健康新西兰兔75只,按随机数字表法分为5组:桃红四物汤低、中、高剂量组、模型组、假手术组,每组15只。假手术组不进行皮片移植,切开后直接缝合,术后予蒸馏水灌胃,桃红四物汤低、中、高剂量组造模后予以相应剂量的桃红四物汤灌胃,模型组造模后以蒸馏水灌胃,各组灌胃时间最长均为8 d。分别于造模后2 d、5 d、8 d从每组随机抽取5只,检测移植皮片血管密度及血清中MMP-2含量。结果:造模后2 d、5 d、8 d,桃红四物汤低、中、高剂量组血管密度、血清MMP-2浓度均高于模型组(P<0.05);造模后2 d桃红四物汤低、中、高剂量组与模型组血管密度均低于假手术组(P<0.05);造模后2 d、5 d桃红四物汤中、高剂量组血管密度均高于桃红四物汤低剂量组(P<0.05)。造模后2 d、5 d、8 d桃红四物汤低、中、高剂量组血清MMP-2浓度均高于假手术组(P<0.05);造模后2 d、5 d、8 d桃红四物汤中、高剂量组血清中MMP-2浓度均高于桃红四物汤低剂量组(P<0.05);桃红四物汤高、中剂量组血管密度、血清MMP-2浓度比较,差异均无统计学意义(P>0.05)。结论:桃红四物汤能上调自体皮片移植模型兔血清中MMP-2含量,并能使血管密度值增加,具有促血管新生作用。Objective: To observe the effect of Taohong Siwu Decoction (桃红四物汤, THSWD) on mi- crovessel density, MMP-2 after rabbit skin graft, and to explore the mechanism of promoting angiogenesis of THSWD. Methods: The 75 healthy New Zealand rabbits were randomly divided into 5 groups: THSWD low dose group, middle dose group and high dose group, model group, sham operation group, 15 rats in each group. Sham operation group without skin grafting, just sutured directly after the incision, and distilled water, while THSWD low dose group, middle dose group and high dose group after modeling was given THSWD by gavage, and model group was treated with distilled water, in each group for the longest time to 8 days. After modeling 2 days, 5 days and 8 days, 5 rabbits were respectively selected to observe the vascular density and MMP-2. Results: The vascular density and MMP-2 of THSWD high, middle, low dose group were higher than the model group after modeling 2 days, 5 days and 8 days (P〈0.05). The vascular density of THSWD high, middle, low dose group and model group were lower the sham operation group after modeling 2 days (P〈0.05). The vascular density of THSWD high, middle dose group were higher than the low dose group after modeling 2 days and 5 days (P〈0.05). The serum MMP-2 concentration of THSWD high, middle, low dose group were higher than the sham operation group after modeling 2 days, 5 days and 8 days (P〈0.05). The serum MMP-2 concentration of THSWD high, mid- die dose group were higher than the low dose group after modeling 2 days, 5 days 8 days (P〈0.05). The vascular density and MMP-2 in THSWD high, middle dose group had no no significant difference (P〉0.05). Conclusion: THSWD can increase the serum MMP-2 concentration and vascular density, to promote angiogenesis.
