摘 要:本试验通过研究TEKT4和TEKT5基因在鸡精子抗冻特性中发挥的生物学功能,为今后在家禽产业中改进鸡精液冷冻效果、提高其冷冻保护率奠定理论基础。随机抽取30只海兰褐种公鸡,收集鸡精液样本并检测质量。采用RT-PCR方法检测冷冻前、后TEKT4和TEKT5基因的表达水平。结果表明,在鸡精子冷冻后TEKT4和TEKT5的差异倍数分别是0.3481、0.5741,表达量均显著下调。因此,推测TEKT4和TEKT5基因与鸡精液的抗冻性相关,可作为今后鸡精液抗冻性研究的候选基因。In this research, the biological function of TEKT4 and TEKT5 genes in the process of chicken semenfreezing was studied, laying a theoretical foundation for improving the effect of chicken semen cryopreservationin poultry industry in the future. The semen samples were collected from 30 hyline brown roosters randomly,and the quality was detected. Then, RT-PCR was applied to detect the transcriptional differences of TEKT4 and TEKT5 before and after semen freezing. The results showed that the transcriptional level of TEKT4 and TEKT5 in cock sperm was significantly decreased after freezing and the fold change was 0.3481 and 0.5741 respectively. Thus, we conclude that TEKT4 and TEKT5 are related to the anti-freezing characteristics of cocksemen, and could be studied as candidate genes in the future.
