摘  要:目的:建立超高效液相色谱-三重四极杆/线性离子阱质谱(UPLC-QTRAP-MS/MS)同时测定玄参中环烯醚萜苷、苯丙素苷及有机酸类12个功效成分的方法,并对其多元功效成分的动态积累进行分析与评价。方法:采用BDS HYPERSIL C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-水为流动相,梯度洗脱,负离子多反应监测(MRM)模式测定;用灰色关联度分析结合DTOPSIS法对多元功效成分进行综合评价。结果:12个功效成分的浓度与峰面积的线性关系良好(r〉0.999 0);平均加样回收率为98.8%-101.2%,RSD均小于2%。玄参中各功效成分的积累动态不一,立冬前后采收的玄参药材综合质量较好,与当地传统采收期基本相符。结论:本文为揭示浙玄参中多元功效成分的动态积累规律及确定其药材的适宜采收期提供基础资料,同时为玄参药材内在质量的综合评价提供新的方法参考。Objective: To develop a UPLC-QTRAP-MS/MS method for the determination of iridoid, phenylpropanoid glycosides and organic acids in Scrophulariae Radix at different harvest time, and to analyze and evaluate the dynamic accumulation of the multiple active constituents in Scrophulariae Radix. Methods: Chromatographic separation was conducted on a BDS HYPERSIL C18 (250 mm ×4.6 mm, 5 μm )column with gradient elution using acetonitrile and water as mobile phases. MS analysis was set in electrospray ionization operated in negative mode. Grey related degree and DTOPSIS were used for comprehensive evaluation of the multiple active constituents. Results: The results demonstrated that all twelve active constituents showed goodlinearity ( r〉0. 999 0 ) in the tested concentration range. The average recoveries of the twelve active constituents ranged from 98.8% to 101.2% with relative standard deviations ( RSDs ) less than 2%. There were differences in the contents of twelve active constituents in Scrophulariae Radix at different harvest periods. The results showed that Scrophulariae Radix harvested around the beginning of winter had high quality, which was consistent with the local traditional harvest period. Conclusion: The result revealed the dynamic accumulation pattern of twelve active constituents in Scrophulariae Radix from Zhejiang province and provided the basis for the suitable harvest time. Meanwhile, it provides new methodological reference for the comprehensive evaluation of inherent quality of Serophulariae Radix.
