摘  要:目的通过观察囊胚玻璃化冷冻对复苏移植后小鼠的生长发育、神经功能、行为学和生殖能力的影响,评估该技术的长期安全性。方法通过促排卵收集性成熟期前C57BL/6雌鼠囊胚,玻璃化冷冻,复苏移植到IGR代孕鼠。仔鼠出生后,观察产仔数和有无外观畸形,记录新生鼠不同时期的生理指标;以正常C57BL/6小鼠为对照,通过转轮、水迷宫和社交试验检测小鼠的运动协调、学习记忆和社交习性;8周龄后检测生殖力。结果囊胚玻璃化冷冻鼠发育指标和行为学各项测试结果与对照小鼠对比差异没有统计学意义;囊胚冷冻雄鼠的生殖力(37.5%)和每胎产仔数[(6.7±2.5)只]与对照组[35.0%,(6.5±1.9)只]差异没有统计学意义。结论囊胚玻璃化冷冻出生的小鼠在生长发育、行为学特征和生殖力等方面和对照鼠无明显差异,表明小鼠囊胚玻璃化冷冻技术是安全和可靠的。Objective This study aimed to investigate the potential impact of mouse blastocyst vitrification on offspring growth, nerve function, behavior and fertility to assess the long-term safety of this technology. Methods Premature C57BL/6 female mice were superovulated. Blastocysts were collected, vitrified and transplanted into ICR surrogate mouse. After the pups were born, litter size and abnormalities, if any, were observed. Physiological development indices at different observation points were recorded. Rotarod test, water maze and social behavior test were done to check motion coordination, learning and memory function and social behavior. Fertility test was carried out when male mouse reached 8 weeks of age. Results Our results showed that there were no statistical difference in any of the development indicators, behavior tests and social ability compare to control mice. Blastocyst vitrification male mice fertility and litter size were 37.5 % and 6. 7 ± 2. 5, respectively, comparable to 35.0% and 6. 5 ± 1.9, respectively, in the control group. Condusion Blastocyst vitrification has no impact on mouse growth, behavioral characteristics and male fertility, suggesting that blastocyst vitrification is a safe and reliable technique for embryo eryopreservation.
