摘 要:建立高效液相色谱(HPLC)测定柯里拉京含量的方法,比较不同植物材料中柯里拉京的含量,筛选出柯里拉京含量高的植物,并初步研究柯里拉京对人胃癌细胞增殖的抑制作用.HPLC结果显示:龙眼核、叶下珠全草、龙眼壳、榄仁树叶和纤梗叶下珠全草的柯里拉京含量较高,依次为643,619,569,271和159μg/g.通过体外实验初步研究了柯里拉京对人胃癌细胞生长的抑制作用,噻唑蓝(MTT)法测得柯里拉京对人胃癌SGC7901细胞的半抑制浓度(IC50)为(42.46±3.31)μmol/L;吖啶橙/溴化乙锭(AO/EB)和Hoechst33258染色结果表明,柯里拉京能诱导人胃癌SGC7901细胞凋亡.建立的HPLC检测方法能较快速、准确地检测出不同植物的柯里拉京含量,所测的12种材料中龙眼核的柯里拉京含量最高;柯里拉京具有抑制人胃癌SGC7901细胞增殖的活性,并且能够诱导其凋亡.To filter plants with high corilagin content,a high performance liquid chromatography (HPLC) method was established for determination of corilagin contents in different plants, and the anti-gastric cancer activity of corilagin was also investigated by thi- azolyl blue (MTT) assay, acridine orange/ethidium bromide (AO/EB) staining, and Hoechst 33258 staining. HPLC analyses showed that longan (Dimocarpus longan Lour.) seed,Phyllanthus urinaria L., longan shell, Terminalia catappa L. leaf and P. tenellus Roxb. contained higher levels of corilagin,which were 643,619,569,271 and 159 pg/g respectively.MTT assay results showed that gastric cancer cells SGC7901 had ICso value of (42.46±3.31) μmol/L with corilagin treatment.AO/EB staining and Hoechst33258 staining showed that corilagin induced apoptosis of gastric cancer cells SGC7901.As a conclusion,the conditions of HPLC explored in this study can quickly and accurately detect corilagin content of different plants, and longan seed contains the highest level of corila- gin among the 12 kinds of plant samples,and corilagin exhibits, anti-gastric cancer activity.
