摘 要:【目的】探索小麦新种质P13对条锈病的抗病性及其抗性机制,为P13种质的进一步推广利用提供理论依据。【方法】以铭贤169为感病对照,用6个条锈菌生理小种(CYR23、CYR29、CYR31、CYR32、CYR33和Hybrid46-8)对P13进行苗期抗条锈性鉴定;采用自然诱发法鉴定P13的田间抗条锈性;以P13和铭贤169为材料,采用常规的夏孢子悬浮液涂抹法接种条锈菌CYR29,以不接种为对照,然后测定多酚氧化酶(PPO)、过氧化物酶(POD)、苯丙氨酸解氨酶(PAL)、β-1,3-葡聚糖酶和几丁质酶等抗病相关酶活性。【结果】苗期P13对CYR32表现为免疫,对CYR29、CYR33和Hybrid46-8表现为近免疫,对CYR23表现为高抗,对CYR31表现为感病。P13在拔节期、孕穗期和乳熟期均表现为免疫或近免疫。P13接种条锈病菌CYR29后,其叶片内POD、PAL、PPO、β-1,3-葡聚糖酶和几丁质酶的活性均高于未接种植株和感病对照铭贤169。【结论】P13对贵州小麦条锈菌生理小种表现出较强的抗病性,其在贵州小麦生产及抗病育种方面具有极高的应用价值。[Objective ]The stripe rust resistance and resistance mechanism of new wheat germplasm P13 were investi- gated, in order to provide theoretical basis for further extension and utilization. [MethodlThe resistance of P13 to six physiological races of Puccinia striiformis f. sp. tritici(CYR23, CYR29, CYR31, CYR32, CYR33 and Hybrid46-8)was identified at seedling stage, with Mingxian 169 as susceptible control. The field resistance of P13 to stripe rust was identi- fied through natural induction. Furthermore, P13 and Mingxian 169 were inoculated with CYR29 by smearing uredospore suspension, with non-inoculation as control, then the activities of polyphenol oxidase (PPO), peroxidase (POD), pheny- lalanine ammonia-lyase(PAL), 13-1,3 glucanase and chitinase were determined by spectrophotometry. [Result]The re- sults showed that, P13 at the seedling stage was immune to CYR32, and nearly immune to CYR29, CYR33 and Hy- brid46-8. Moreover, P13 was highly resistant to CYR23, and susceptible to CYR31. At the jointing stage, booting stage and milky-ripe stage, P13 was immune or nearly immune to all stripe rust races. In addition, the activities of POD, PAL, PPO, 13-1,3-glucanase and chitinase of P13 inoculated with CYR29 were higher than those of uninoculated wheat and susceptible control(Mingxian 169). [Conclusion]P13 shows high resistance to local wheat stripe rust races in Guizhou, so it has very high application value in wheat production and breeding for disease-resistance.
