不同剂量盐酸氨溴索对伏立康唑在小鼠血浆及肺组织中分布的影响

摘　　要：目的:研究不同剂量盐酸氨溴索对伏立康唑在小鼠血浆及肺组织中分布的影响。方法:将240只昆明雄性小鼠,随机分为对照组和实验组,其中实验组包括低、中、高3个剂量组。实验动物均灌服伏立康唑66.67 mg·kg^(-1),各实验组则分别加服不同剂量盐酸氨溴索(分别为10,50,150 mg·kg^(-1))。于给药后不同时间点摘眼球取血并采集肺部组织。使用高效液相色谱法测定血浆及肺组织中伏立康唑浓度。以DAS 2.1.1软件拟合药动学参数,并使用SPSS 21.0软件对4组药动学参数进行统计学分析。结果:血浆及肺组织中伏立康唑的主要药动学参数AUC0-t、AUC0-∞、t1/2、tmax、CL、V、Cmax,在4组间均无显著差异(P>0.05)。结论:盐酸氨溴索对伏立康唑在小鼠血浆及肺组织中分布无影响。OBJECTIVE To study the effects of different doses of ambroxol on distribution of voriconazole in plasma and lung of mice.METHODS Two hundred and forty male Kunming mice were randomly divided into control group and experimental group.Experimental group was further divided into high,medium and low dose groups.All mice were orally administrated with voriconazole（66.67 mg·kg-1）as control group.In three experimental groups,low（10 mg·kg-1）,medium（50mg·kg-1）and large（150 mg·kg-1）doses of ambroxol were added,respectively.Blood was sampled by removing eye ball at different times.Then mice were decapitated immediately and lung samples were obtained.Drug mass concentrations in plasma and lung were determined by high performance liquid chromatography method.Pharmacokinetic parameters of four groups were calculated with DAS 2.1.1 program software and statistically analyzed with SPSS21.0software.RESULTS Main pharmacokinetic parameters such as AUC0-t,AUC0-∞,t1/2,tmax,CL,V,Cmaxin plasma and lung of four groups had no significant differences（P〉0.05）.CONCLUSION Distributions of voriconazole both in plasma and lung have not been affected by different doses of ambroxol.
