摘  要:目的:研究相思藤水提物(XSTE)对小鼠化学性及酒精性肝损伤的保护作用及机制。方法:分别采用四氯化碳(CCl4)、对乙酰氨基酚(AP)腹腔注射小鼠,建立化学性肝损伤模型;用50%酒精灌胃小鼠建立酒精性肝损伤模型。将昆明种小鼠随机分为正常对照组、肝损伤模型组、联苯双酯(BPDC)阳性对照组、XSTE高、中、低剂量组。计算肝脏重量指数和脾脏重量指数,测定血清谷草转氨酶(AST)、谷丙转氨酶(ALT)活性,肝组织丙二醛(MDA)和谷胱甘肽(GSH)含量、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性,酒精性肝损伤研究测定血清乙醇浓度、抗利尿激素(ADH)和乙醛脱氢酶(ALDH)。结果:与模型组比较,XSTE能减轻肝脏和脾脏重量,降低血清AST、ALT活性,降低血中乙醇浓度,增加ADH和ALDH活性,降低肝组织MDA含量,增加肝组织SOD、GSH-Px活性,增加GSH含量(P<0.05或P<0.01)。结论:XSTE对小鼠化学性及酒精性肝损伤具有明显的保护作用,其机制可能与其抗氧自由基、抑制脂质过氧化作用有关。Objective:To investigate the effect and mechanism of Xiang Si Teng aqueous extract(XSTE)on CCl4-,acetaminophen(AP)-and alcoholic-induced acute liver injuries in mice.Methods:The mice were given by gavage with 50%ethanol,and by intraperitoneal injection with CCl4 or AP to establish models of alcoholic liver injury and chemical liver injuries,respectively.The mice were randomized into 6groups:normal control,model control,positive control,low-,medium-and high-dose XSTE-treated groups.The liver and spleen indexes were calculated.The activities of ALT and AST in the serum,and the levels of SOD,MDA,GSH-Px and GSH in liver tissue were measured.The alcohol concentration and the activities of ADH and ALDH were also detected in the study of alcoholic liver injury.Results:Compared with the model group,XSTE significantly reduced indexes of liver and spleen as well as the levels of MDA,AST and ALT,while increased the levels of SOD,GSH and GSH-Px significantly(P〈0.05 or P〈0.01).It also reduced the alcohol concentration and the activities of ADH and ALDH.Conclusion:XSTE could protect the mice from CCl4-and AP-induced acute chemical liver injuries and attenuate alcoholic liver injury,and its mechanisms may be related to its effect in attenuating free radical and inhibiting the lipid peroxidation.
