摘 要:目的:探讨镉(Cd)对孕鼠的生殖毒性及胎鼠的发育毒性作用。方法:将受孕Sprague Dawley(SD)大鼠随机分成氯化镉(CdCl2)组(Cd组)和对照组(C组)。Cd组孕鼠每天饮用含40mg/L CdCl2的蒸馏水,C组饮用等量蒸馏水,均用一般饲料喂养。取20d的孕鼠麻醉,心脏取血,测定血清中Cd含量。每组随机取20d的孕鼠10只,麻醉后剖宫取出胎盘组织,称重,切取约1mm×1mm×1mm的胎盘组织块于4%戊二醛中固定,电镜下观察胎盘超微结构。每组随机取20只胎鼠,准确称重,测定体、尾长,取胎盘及胎鼠肝脏分别称重,测定其组织Cd含量。结果:与C组比较,Cd组孕鼠血清Cd[(0.069±0.015)μmol/L vs(0.023±0.009)μmol/L]、胎盘Cd[(0.382±0.071)μg/g vs(0.143±0.022)μg/g]及胎鼠肝脏Cd[(18.088±5.788)ng/g vs(9.075±4.428)ng/g]含量均明显升高(均P<0.05)。与C组相比,电镜下可见Cd组胎盘细胞结构被严重破坏。与C组相比,Cd组胎盘重[(0.46±0.09)g vs(0.64±0.10)g],胎鼠体重[(5.33±0.40)g vs(6.12±0.41)g],胎鼠体长[(48.55±1.76)mm vs(51.51±1.84)mm],胎鼠尾长[(14.84±1.23)mm vs(16.67±1.69)mm]均显著降低(均P<0.01)。结论:Cd对哺乳动物及其后代有毒性作用,Cd接触是妊娠期间的一个危险因子。Objective:To investigate the reproductive and embryos' developmental toxicities of cadmium in pregnant rats.Methods:Sprague Dawley(SD)pregnant rats during the same period were randomized into the cadmium chloride group(Cd group)and the control group(CN group).The Cd group mice drank the water containing cadmium chloride of 40mg/L per day,the CN group drank the same amount of distilled water.Both groups of rats were fed with a normal diet.On day 20 of gestation,10 pregnant rats from both groups were randomly selected and anesthetized,then cardiac blood was drawn to detect the cadmium content in serum,and the cesarean section was performed to take out the placenta for weighting and giving ultrastructural analysis.The fetuses from both groups were examined carefully,including rat body weight,the length of body and tail,and the cadmium content in the liver.Results:Compared with the CN group,the serum cadmium,placental cadmium and fetal rat liver cadmium were significantly increased in the Cd group(P〈0.01).The cell structure is destroyed in the pregnant rats placenta of Cd group,as showed with electron microscope.The placental weight,fetal weight and the body and tail length of Cd group were significantly lower than those of the CN group(P〈0.05).Conclusion:Cadmium can cause toxicity to reproductive system of mammals,cadmium exposure is a risk factor in pregnancy.
