摘  要:目的:探讨p53凋亡刺激蛋白2(ASPP2)DNA甲基化在老年大鼠肾基脏底膜增厚中的作用和机制。方法:以6月龄大鼠为成年组,18月龄大鼠为老年组,应用过碘酸希夫(PAS)染色,光镜下观察各组大鼠肾脏基底膜改变,qRT-PCR、Western-blot分别检测各组大鼠ASPP2 mRNA及蛋白表达,n MS-PCR法检测ASPP2甲基化水平,qRT-PCR检测DNMT1 mRNA表达改变。结果 :老年大鼠较成年大鼠肾脏基底膜明显增厚,ASPP2 mRNA及蛋白的表达在老年组大鼠中显著增高(P<0.01);与成年组相比,老年组ASPP2 DNA甲基化水平显著下降(P<0.01),且DNMT1 mRNA及蛋白表达也呈下降改变(P<0.01)。结论:ASPP2 DNA甲基化是促进老年大鼠肾脏基底膜增厚的重要机制,DNMT1参与了其调控。Objective To explore the effect of DNA methylation of apoptosis stimulating protein 2 of p53 (ASPP2) gene on glomerular basement-membrane thickening in old rats. Methods Sprague-Dalwley (SD) rats were randomly divided into two groups: six-month old rats were assigned to the adult group (group N, n = 12), eighteen-month old rats were assigned to the elderly group (group O, n = 12). The change of glomerular basement- membrane of each group was detected by PAS staining assay. The protein and mRNA expression of ASPP2 in the kidney were detected by western blotting and real time polymerase chain reaction (RT-PCR). The level of ASPP2 methylation and DNMT1 expression were detected by nMS-PCR and qRT-PCR, respectively. Results Compared with the adult group, the glomerular basement-membrane thickening and ASPP2 expression were significantly increased (P 〈 0.01), but the DNA methylation level of ASPP2 gene and DNMT1 mRNA expression were significantly decreased in the elderly group (P 〈 0.01). Conclusions The DNA methylation of ASPP2 is an important mechanism underlying the glomerular basement-membrane thickening in elderly rats, as well as the involvement of DNMT1 expression.
