摘  要:目的:研究2型糖尿病患者的唾液蛋白质组,筛选差异蛋白质指纹图谱,建立2型糖尿病的蛋白质指纹图谱诊断模型。方法:采集2型糖尿病患者70例和健康体检者45例的唾液标本,采用WCX纳米磁珠联合基质辅助激光解析离子化飞行时间质谱(matrix assisted laser desorption ionization time-of-flight mass spectrometry,MALDI-TOF-MS)技术检测,筛选两组间特异性表达差异蛋白,并结合生物信息学方法建立诊断模型。结果:共获得158个差异有统计学意义的蛋白质峰(P<0.05)。选择质荷比(m/z)为3447.57 Da、4735.24 Da两个差异蛋白质峰构建最佳决策树模型,该模型的准确率为89.57%(103/115),敏感性91.4%(64/70),特异性为86.7%(39/45)。结论 :初步建立了2型糖尿病唾液蛋白质组诊断模型,为其早期诊断提供了新的方法和途径,具有重要的临床诊断意义。Objective To develop a protein-molecule diagnostic model for diabetes mellitus type 2 through proteomic technologies to separate and identify the protein mapping with significance deviation of patients with diabetes mellitus type 2. Methods Saliva samples from 70 patients with diabetes mellitus type 2 and 45 healthy volunteers were purified by WCX and MALDI-TOF-MS, then the specifically expressed proteins in two groups were used to establish the diagnostic model through bioinformatic methods. Results The saliva protein fingerprint data of the two groups were compared and 158 discrepant protein peaks were found (P 〈 0.05 ). The 2 peaks of M/Z 3447.57 Da and 4735.24 Da were used to build the best decision tree model. The accuracy, sensitivity and specificity of the diagnostic model were 89.57% ( 103/115 ), 91.4% (64/70), and 86.7% (39/45) respectively. Conclusion Preliminary saliva proteome diagnosis model is set up, providing a new way for early diagnosis of type 2 diabetes mellitus.
