摘 要:油桐作为我国四大木本油料植物之一,其主产物桐油广泛应用于工业领域,极具经济价值和开发前景。为研究油桐种仁在不同发育时期其乙酰辅酶A羧化酶活性与含油率的变化及其二者之间的相关性,本文以3个不同家系油桐种仁为试验材料,采用索氏抽提法和植物乙酰辅酶A羧化酶酶联免疫分析试剂盒(ELISA)测定其8个发育时期的含油率与乙酰辅酶A羧化酶活性,并进行相关性分析。结果表明:1)不同家系油桐种仁中的含油率在各个不同发育时期均存在极显著差异(P<0.01);2)8个发育时期的油桐种仁含油率在供试的3个油桐家系中均表现出S型走势,即前期积累速度较慢,中间积累速度较快,后期积累速度又放缓,进入平台期;3)3个油桐家系的ACCase活性在8个时期均表现出双峰的波浪型走势,第1个峰值出现在Ⅱ-2时期,第2个峰值出现在Ⅲ-2时期;(4)含油率与ACCase活性在3个不同家系中均表现出一定的相关性,其中9号家系为极显著的正相关关系,其余表现出显著的正相关关系。该结果表明可以通过测定油桐种仁中ACCase活性来侧面反映其含油量的高低。As a famous kind of wood oil trees in China, the seeds of Vernicia fordii can used to producing tung oils which have a widely use in industries. In order to analyze the change of Acetyl-Co A carboxylase activity and oils content and their correlation during the different development periods. The seeds from three different families of V. fordii were picked out as experimental materials in this paper, and the method of Soxhlet extraction and ELISA kit were used measure the activity of acetyl-Co A carboxylase and oils content at eight growth stages and the relativity of them was analyzed as well. The results were as follows:(1) There exist a very significant difference in the oils content in the different development periods in all tested V. fordii families(P 0.01).(2) The dynamic changes of oils content in all tested V. fordii families at eight developmental stages showed "S"-type movements. It means that the speed of oils accumulation was slow in the beginning;intermediate accumulation speed was fast, the late accumulation rate slowed down again, and got into the platform.(3) The dynamic changes of ACCase acitivity in three different families at eight periods showed a double peak wave type trend, the first peak appears in the second-2 period(Ⅱ-2), the second peak appears in the third-2 period(Ⅲ-2).(4) The oils content and ACCase activity in three different families showed a certain correlation. The family(No. 9) extremely significant positive correlation, and the rest showed significant positive correlation. This result showed that we can be measured by Tung kernel ACCase activity to reflect the content of the fatty acid level.
