摘 要:目的优选复方榆树中咖啡酸的提取工艺。方法采用HPLC测定复方榆树中指标成分咖啡酸转移率,流动相:甲醇-0.4%磷酸(27∶73);色谱柱:Agilent Eclipse Plus C_(18)(4.6μm×250μm,5μm);柱温:30℃;检测波长:323 nm;检测器:二极管阵列检测器(DAD);分析时间:25 min;流速:1.0 m L·min^(-1);进样量:10μL。以咖啡酸的转移率为指标,通过单因素试验考察提取方法、料液比、提取时间、提取次数、粉碎度对提取工艺的影响,并采用正交试验优化提取工艺。结果最佳提取工艺为复方药材过60目筛网,用8倍量60%乙醇加热回流提取3次,每次30 min。结论优选的提取工艺稳定可行,为复方榆树的开发研究提供参考。Objective To optimize the extraction process of caffeic acid from Chinese medicine compound Ulmaceae.Methods HPLC was employed to determine the transfer rate of caffeic acid in compound Ulmaceae.The analysis was performed on Agilent Eclipse Plus C18column(4.6×250,5 μm) at 30 ℃ with DAD detector.The mobile phase consisted of methanol-0.4 % phosphoric acid(27 ∶73).The detection wavelength was 323 nm;the analysis time lasted for 25 min;the flow rate was 1.0 ml·min^-1and the injection volume was 10 μL.With caffeic acid transfer rate as the observation index,single-factor experiment was used for the investigation of effects of extraction method,solid-liquid ratio,extraction time,extraction times,particle size of compound Ulmaceae on the extraction process,and then we optimized the extraction process by orthogonal experiment.Results The optimum extraction technology of compound Ulmaceae was as follows:medicinal particle size was able to go through 60-mesh screen after grinding;the medicine was processed with heating reflux extraction with 8 times volume of 60 % ethanol for 3 times,30 min for each time.Conclusion The optimal extraction process is stable and feasible,and can provide reference for the development and research of compound Ulmaceae.
