白桦脂醇拮抗地塞米松诱导成骨细胞内脂质积聚影响细胞凋亡的实验研究

摘　　要：目的观察白桦脂醇和地塞米松共同作用于小鼠MC3T3-E1细胞后,内源性脂质蓄积与细胞凋亡中的关系。方法以MC3T3-E1细胞为研究对象,采用白桦脂醇和地塞米松共同作用于MC3T3-E1细胞,测定细胞内甘油三酯含量;NileRed荧光染色观察脂质蓄积情况;流式细胞技术测定成骨细胞凋亡率;ELISA法检测Caspase-3酶活性;采用Spearman等级相关进行脂质含量与成骨细胞凋亡相关性分析。结果白桦脂醇和地塞米松共同作用于小鼠MC3T3-E1细胞后,0.5μmol/L地塞米松(B组)与1.0μmol/L地塞米松组(C组)较空白组(A组),细胞凋亡率、Caspase-3酶活性均明显增高(P<0.05),细胞内甘油三酯含量明显增加。2.0μg/ml白桦脂醇作用细胞后,地塞米松所致的细胞凋亡率上升与Caspase-3酶活性升高受到明显抑制,同时细胞内的甘油三酯蓄积降低。Spearman等级相关分析表明地塞米松诱导后的成骨细胞内脂质含量与细胞凋亡率呈正相关,r=0.412,P<0.05。结论白桦脂醇可显著减轻地塞米松诱导的成骨细胞内脂质蓄积,降低细胞凋亡率;提示地塞米松所致细胞凋亡至少部分与细胞内脂质蓄积有关。Objective To observe the correlation between endogenous lipid accumulation and apoptosis of MC3T3-E1 cells affected by betulin and dexamethasone in vitro. Me thods MC3T3-E1 cells were used as the research subject. The cells were treated with betulin and dexamethasone. The lipid synthesis in the cells was determined with triglyceride content examination and Nile redstaining. The apoptosis rate of osteoblasts was detected with flowcytometry. Caspase-3 enzyme activity was detected with ELISA. The correlation between lipid content and cell apoptosis was analyzed with Spearman rank method. Re sults The cell apoptosis rate,Caspase-3 activity,and the lipid content in the cells increased significantly in cells affected by 0. 5 μmol / L（ group B） and 1 μmol / L dexamethasone（ group C） than in control group（ P〈0. 05）. Betulin significantly decreased the apoptosis rate and the Caspase-3 activity,as well as the lipid accumulation in the cells. Spearman rank correlation analysis indicated that the lipid content was positively correlated with the rate of apoptosis of osteoblasts（ P〈0. 05,r = 0. 412）. Conclusion Betulin significantly reduces the dexamethasone-induced apoptosis of osteoblasts and lipid accumulation,suggesting that hormone induced apoptosis is at least in part correlated with the intracellular lipid accumulation.
