摘  要:目的 优选虫草多糖脂质体制备的最佳工艺。方法 采用逆向蒸发法制备发酵虫草菌粉多糖脂质体,以包封率为指标,以药脂比、卵胆比与水油比为因素,采用L9(34)正交试验设计对制备条件进行优选,鱼精蛋白测定发酵虫草菌粉多糖脂质体的包封率,透射电镜及马尔文激光粒度分析仪测定其形态和粒径。结果 逆向蒸发法制备发酵虫草菌粉多糖脂质体的最佳工艺为大豆卵磷脂与药物的比例为20∶1,大豆卵磷脂与胆固醇的比例为2∶1,乙醚与磷酸盐缓冲液的比例为3∶1。结论 采用最佳工艺制备的发酵虫草菌粉多糖脂质体包封率较高,形态和粒径较均匀,重现性好。OBJECTIVE To optimize the preparation conditions of Polysaccharides fromfermented cordyceps liposome. METHODS Polysaceharides fromfermented cordyceps liposome was prepared by reverse evaporation method. L9 ( 34 ) orthogonal test wan adopted to optimize the preparation conditions which was designed with three factors of lecithin to drug ratio, lecithin to cholesterol ratio, and water to oil ratio, and index of encapsulation efficiency. The encapsulation efficiency of Polysaecharides fromfermented cordyceps liposome was assayed by protamine method, the shape and particle size were observed by transmission electron microscope. RESULTS The optimized preparation conditions were as follows:The ratio of lecithin to drug was 20:1 ,the ratio of lecithin to cholesterol was 2:1 ,and water-oil ratio was 3: 1. CONCLUSION The Polysacchafides fromfermented cordyceps liposome prepared under the optimized conditions has following advantages: high encapsulation efficiency, uniform shape and particle size, and good reproduction quality.
