改良肝浸液培养基用于布氏杆菌增菌及分离培养的实验研究

摘　　要：目的研究一种专门用于布氏杆菌增菌和分离培养的培养基组分。方法以肝浸液为基础,加入氯化钠,蛋白眎、胨,酵母浸粉,葡萄糖,硝酸镧,赤藓醇,甘油和血清,命之为改良肝浸液,用于布氏杆菌增菌。加入琼脂即为改良肝浸液琼脂,用于布氏杆菌分离。并与血琼脂相比较。结果改良肝浸液培养基的配方:猪肝500 g,蛋白眎、胨5 g,酵母浸粉5 g,葡萄糖8 g,硝酸镧2 g,赤藓醇4 g,甘油20 ml,氯化钠5 g,血清(无布氏杆菌凝集素)20 ml,余量加蒸馏水至1 000 ml。结论改良肝浸液及改良肝浸液琼脂是良好的布氏杆菌增菌和分离培养基。Objective To study a special culture components for the enrichment and isolation of Brucella bacteria. Methods On the basis of liver extracts,adding sodium chloride,peptone,yeast extract powder,glucose,erythritol,glycerol and serum,to name it modified liver extracts in order to enrich Brucella bacteria. With the addition of agar,it was named modified liver extract agar in order to isolate Brucella bacteria. Then to compare it with blood agar. Results To improve the formulation of the modified liver extracts medium： pig＇s liver 500 g,peptone 5 g,yeast extract powder 5 g,glucose 8 g,erythritol 4 g,glycerol 20 ml,sodium chloride 5 g,serum（ no Brucella agglutinin） 20 ml,adding distilled water to 1 000 ml. Conclusion Modified liver extracts and modified liver extract agar is good medium for the enrichment and isolation of Brucella bacteria.
