摘 要:目的建立了用HPLC法对白毛夏枯草中木犀草素、芹菜素同时定量的方法。方法高效液相色谱法,色谱柱BIOSILU546250-1型色谱柱(4.6mm×250mm,5μm);流动相:乙腈-0.4%甲酸水(45:55);检测波长336mm;柱温30℃;流速1.0mgmin;进样量20μl。结果白毛夏枯草木犀草素、芹菜素的浓度与峰面积,分别在8.2900-74.6640μg(r=0.9995)、3.1404-28.2636μg(r=0.9998)范围内呈良好的线性关系;平均加样回收率分别为99.18%、98.63%,RSD分别为1.20%、1.31%。结论3、4月份白毛夏枯草药材中的木犀草素与芹菜素含量明显高于其他几个月份。因为3、4月份为白毛夏枯草的开花期,故推测木犀草素与芹菜素广泛存在于开花期的白毛夏枯草植株中。本方法适合同时测定白毛夏枯草样品中木犀草素与芹菜素成分的含量,分离度好,快速,简便,重现性好。Objective A reserved-phase HPLC method was developed for the determination of luteolin, apigenin in root ofAjuga decumbens Thunb. Method BIOSIL U546250-1-C18 column (4.6mm×250mm,5μm) was used with the mobile phase of acetonitrile -0.4% formic acid: water 45 : 55. The flow rate was at 1.0 ml/ min. The detection wavelength was 336 nm at 30℃. Results The linear response ranged from 8.290 0 - 74.664 0 μg for luteolin (r= 0.999 5, n = 5), 3.140 4 - 28.263 6 μg for apigenin (r = 0.999 85, n = 5). The average of recoveries were 99:18% with RSD 1.20% for luteolin, 98.63% with RSD 1.31% for apigenin. Conclusion The results show that the max content of flavornoid in A.decumbens is March and April, The content of luteolin component is higher in north areas than in south areas of Anhui province. The method is quick, simple and repeatable for determination of luteolin and apigenin in root ofAjuga decumbens Thunb.
