摘  要:目的考察毛冬青浸膏外用对糖尿病溃疡模型小鼠创面愈合率的影响,并初步探讨其可能的作用机理。方法采用链脲佐菌素结合高脂饲料诱导2型糖尿病模型,在此基础上复制皮肤溃疡模型,将模型小鼠随机分为胰岛素组(I),胰岛素+毛冬青低、中、高剂量(I+L,I+M,I+H)组和胰岛素+羟苯磺酸钙(I+CDC)组,分别给药;另取正常小鼠复制皮肤溃疡模型,涂抹等量生理盐水。以面积法测定各组的创面愈合率,并采用免疫组化法检测增殖细胞核抗原(PCNA)和第八因子相关抗原(FⅧ-RAg)的表达情况。结果给药结束后,与单纯溃疡组小鼠相比较,其他各组创面愈合率均显著降低,PCNA的阳性表达率均显著下调;除胰岛素+羟苯磺酸钙组外其余各组FⅧ-RAg的阳性表达率均显著下调;与单用胰岛素组比较,胰岛素联合毛冬青低、中、高剂量(I+L,I+M,I+H)组创面愈合率均显著增加,PCNA和FⅧ-RAg的阳性表达率显著升高。结论毛冬青外用对糖尿病溃疡具有较显著的修复作用,其作用可能与创面组织中PCNA和FⅧ-RAg的表达增加,从而促进表皮细胞增殖和新生血管的形成有关。OBJECTIVE To investigate the effect and mechanism of llex pubescens extractum on wood regeneration of diabetic ulcer rats. METHODS Streptozotocin (STZ) and high-fat diet were used to establish the type 2 diabetic mice model,then skin ulcer model based on diabetic mice model was copyed. Diabetic ulcer rats were divided into insulin group,insulin combine with low, medium, high dose of llex pubescens extractum group (I+L, I+M, I+H group) and insulin combine with Calcium Dobesilate Capsules group (I+ CDC group). Normal rats was used to copy skin ulcer model and given normal saline. Area method was used to measure the wound regeneration and immunohistochemical method was used to detected the positive expression of PCNA and FⅧ- RAg. RESULTS Comparing with the skin ulcer mice, wound regeneration and positive expression of PCNA decreased significantly in other groups, positive expression of FⅧ- RAg was significantly lower except the I+ CDC group. Comparing with the insulin group, wound regeneration and positive expression of PCNA and FⅧ- RAg increased significantly in I+M group and I+H group. CONCLUSION External use of llex pubescens extractum showed a good effect on wound regeneration of diabetic ulcer mice. Increasing of positive expression of PCNA and FⅧ- RAg so as to promote the epidermal cell proliferation and the neovascularization may be one of the mechanisms of llex pubescens extractumon wound regeneration of diabetic ulcer mice.
