摘  要:目的:基于NF-κB信号通路观察小青龙汤合玉屏风散对变应性鼻炎大鼠鼻黏膜IL-6含量和IL-6 m RNA表达的影响,揭示小青龙汤合玉屏风散治疗AR的作用机制。方法:将清洁级健康雄性Wistar大鼠48只将大鼠按体重采用随机区组法分为6组,即分成正常组、AR模型组、Forskolin(c AMP激活剂)干预组、H89(PKA抑制剂)干预组、PDTC(NF-κB抑制剂)干预组、小青龙汤组合玉屏风散组,每组8只。采用卵蛋白全身致敏与局部攻击方法制作AR大鼠模型,观察小青龙汤合玉屏风散组治疗后鼻黏膜IL-6含量和IL-6 m RNA表达,结果进行统计学分析。结果:模型组IL-6含量和IL-6 m RNA表达上调,c AMP激活剂Forskolin干预组、NF-κB抑制剂PDTC干预组、小青龙汤组合玉屏风散治疗组均能使IL-6含量和IL-6 m RNA表达下调,而PKA抑制剂H89能上调IL-6含量和IL-6 m RNA表达。结论:小青龙汤合玉屏风散治疗变应性鼻炎作用机制可能抑制NF-κB信号通路和兴奋c AMP-PKA信号通路使IL-6含量和IL-6 m RNA表达下调有关。Objective:To observe the effect of the Xiaoqinglong Decoction and Yupingfeng Power on the content of IL-6 and the expression of IL-6 m RNA in nasal mucosa of allergic rhinitis rats by NF-κB signaling pathway,and reveal the mechanism of the Xiaoqinglong Decoction and Yupingfeng Power treating AR. Methods:48 healthy male wistar rats were divided into 6 groups by randomized bocks design:normal group,AR model group,Forskolin(c AMP activator)group,H89(PKA inhibitor)group,PDTC(NF-κB inhibitor),and the Xiaoqinglong Decoction and Yupingfeng Power group,8 rats in each group. The AR rat model was made by the method of whole body sensitization and local attack. The content of IL-6 and the expression of IL-6 m RNA was observed after the treatment of the Xiaoqinglong Decoction and YupingfengPower group. The results were statistically analyzed. Results:The content of IL-6 and the expression of IL-6 m RNA in the model group increased,and the content of IL-6 and the expression of IL-6 m RNA in the Forskolin group,PDTC group,the Xiaoqinglong Decoction and Yupingfeng Power group decreased,while the PKA inhibitor H89 could increase the content of IL-6 and the expression of IL-6 m RNA. Conclusion:The mechanism of the treatment of allergic rhinitis by the Xiaoqinglong Decoction and Yupingfeng Power group may inhibit the content of IL-6 and the expression of IL-6 m RNA by inhibition of NF-κB signaling pathway and excitatory c AMP-PKA signaling pathway.
