血清miRNA-208和β-肌球蛋白重链在心力衰竭患者中的表达及其意义

摘　　要：目的检测微小RNA（miRNA）-208及β-肌球蛋白重链（β-MHC）在心力衰竭患者血清中的表达变化及相互关系。方法收集因心力衰竭入院的患者60例（心力衰竭组）和健康体检者56例（对照组），应用实时荧光定量PCR法检测血清miRNA-208、β-MHC的表达，超声心动图评估左室质量指数（LVMI）、左室射血分数（LVEF）情况。结果心力衰竭组血清中miRNA-208、β-MHC表达水平明显升高，差异有统计学意义（P〈0．01）。miRNA-208表达水平与LVMI呈正相关（P〈0．05），与LVEF呈负相关（P〈0．05）。β-MHC与miRNA-208表达水平呈正相关（P〈0．05）。结论心力衰竭患者miRNA-208表达增高及相应的正向上调β-MHC水平可能是心力衰竭心肌重构的机制之一，与心力衰竭的发生发展有关。Objective To investigate change and relationship of microRNA-208 （ miRNA-208 ） and β myosin heavy chain （ β -MHC ） expressions in patients with heart failure. Method There were 60 patients being definitely diagnosed heart failure and 56 healthy controls to be selected. Quantitative real time polymerase chain reaction was applied to assess expressions of miRNA-208 and β-MHC. Transthoracic echocardiographic examination were performed to assess the left ventricular mass index （ LVMI ） and the left ventricular ejection fraction （ LVEF ） . Results The expressions of miRNA-208 and β-MHC markedly increased in heart failure Compared with normal subject. The expressions of miRNA-208 and β-MHC were positively correlated with the LVMI （ P〈0.05 ） and negatively with LVEF （ P〈0.05 ） . The expressions of miRNA-208 were positively correlated with the expressions of β-MHC （ P〈0.05 ） . Conclusion Expression of miRNA-208 level was significantly up-regulated in human patients with heart failure, possibly contributing to up-regulation of β -MHC leading to the development of heart failure. These results provide potential new insights into molecular mechanisms of heart failure.
