金银花中有效成分的含量与其质量等级相关性研究

摘　　要：目的:建立金银花中有机酸、木犀草苷含量测定方法,测定不同等级金银花中有机酸及木犀草苷含量。研究金银花有效成分含量与质量等级的相关性,为金银花的质量控制和应用提供参考。方法:采用Di KMA Diamonsil ODS C18色谱柱(4.6 mm×250 mm,5μm),流动相为乙腈、0.025%磷酸、0.3%醋酸溶液梯度洗脱;流速梯度;柱温40℃,检测波长为326 nm和350 nm。结果:不同等级金银花质量差异较大,出现了等外品中绿原酸、木犀草苷含量符合规定,但咖啡酸、异绿原酸A、B、C含量较低的情况。结论:金银花有效成分含量与其质量等级具有相关性。金银花的质量等级是由多种有机酸和木犀草苷含量共同决定的。上市流通药材质量品质是否符合要求,除《中华人民共和国药典》规定有效成分含量应符合规定外,还应引入其他分析项目,如性状、等级、多种有机酸及指纹图谱的检测。文章所建立的方法可以为金银花的控制和应用提供科学依据。Objective ： To establish a method for the determination of organic acids and luteolin in Lonicera Japonica and deter- mine the contents of organic acids and luteolin glycosides in Lonicera Japonica of different grades. To research the correlation between the effective components and their grades and provide a reference for quality control and application of Lonicera Japonica. Methods：DiKMA Diamonsil ODS C lscolumn（4.6 mm × 250 mm,5 μm）was adopted. The mobile phase was acetonitrile （solvent A） -0.025% phosphoric acid,0.3% acetic acid （solvent B） solution in gradient elution mode. The flow rate varies with time. Detect wavelength was 326 nm and 350 nm. The column temperature was at 40 ℃. Results ： Different grades of Lonicera Japonica have different active ingredient contents and there were large differences in the quality of different grades, chlorogenic acid and luteolin contents were compliance in some substandard products, but caffeic acid, chlorogenic acid A, B and C contents lower. Con- clusion ： The active ingredient contents of Lonicera Japonica are relevant to their quality grades. A variety of organic acid contents and luteolin glycosides jointly decide the quality grades of Lonicera Japonica . When we consider the market product of Lonicera Japonica is in accordance with the provisions or not, not only the content of effective components should comply with the Pharma- copoeia, but also other analysis projects should be introduced in, such as characters, grades, a variety of organic acids and fingerprint testing. The results can provide a scientific basis for quality control and application of Lonicera Japonica.
