摘  要:目的:探讨清化颗粒对db/db糖尿病小鼠肠道环磷酸腺苷(c AMP)和三磷酸腺苷(ATP)的影响。方法:6只雄性db/m小鼠为空白对照组,24只雄性db/db糖尿病小鼠随机分为模型对照组、清化颗粒低剂量组、清化颗粒中剂量组和清化颗粒高剂量组,干预4周,IPGTT法观察小鼠血糖水平变化,ELISA法分析小鼠肠道c AMP、ATP和血清胰高血糖素样肽-1(GLP-1)的水平,组织病理学观察小鼠回肠组织形态变化。结果:与模型对照组比较,清化颗粒下调db/db糖尿病小鼠的空腹血糖(FBG)(P<0.01),增加肠道c AMP水平(P<0.01),降低肠道ATP水平(P<0.01)和血清GLP-1水平(P<0.01),并具有剂量依赖性。清化颗粒明显改善db/db糖尿病小鼠回肠组织的形态。结论:清化颗粒能够降低db/db糖尿病小鼠的血糖水平,促进GLP-1分泌,改善回肠组织形态,其机制可能与增加肠道c AMP水平,降低肠道组织中ATP水平有关。Objective:To explore the intervention effect of Qinghua Granule (QHG) on ATP and cAMP in db/db mice ileum tissue. Methods : Six male db/m mice were as normal control group. Twenty - four male db/db mice were randomized into model control group, QHG low - dose group, QHG middle - dose group and QHG high - dose group. All the groups were administrated for 4 weeks. Blood glucose was detected by IPGTT. The serum concentration of GLP - 1 was tested by ELISA. The levels of ATP and cAMP in ileum of db/db mice were detected by ELISA. The structure of ileum tissue was observed by histopathological analysis. Results:Compared with model control group, QHG could decrease FBG and increase GLP - 1 secretion obviously (P 〈 0. 01 ). QHG downregulated the cAMP levels and up- regulated the ATP level in ileum tissue significantly (P 〈 0. 01 ). In addition, QHG obviously improved the structure of ileum of db/db mice. Conclusion : QHG could reduce FBG obviously, improve GLP - 1 secretion and the structure of ileum and the mechanism might be related to the regulation of ATP and cAMP.
