摘  要:目的:观察参知健脑胶囊对拟血管性痴呆大鼠皮层和海马中Glu和GABA的影响。方法:将70只SD大鼠随机分为假手术组、模型组、安理申组、金纳多组以及参知健脑胶囊低、中、高剂量组,每组10只,采用2VO法建立拟血管性痴呆大鼠模型。除假手术组和模型组给予等体积生理盐水灌胃外,其余各组分别给予安理申1 mg/kg、金纳多20mg/kg以及参知健脑胶囊40 mg/kg、120 mg/kg、360 mg/kg灌胃。各组于造模后7 d开始给药,每日1次,连续灌胃8周。本别在术后第5、8周以Morris水迷宫检测其学习记忆能力;给药8周后大鼠统一处死后取皮层、海马,用H&E、Nissl's染色法观察皮层海马的组织形态学改变和神经元存活状况,用高效液相色谱法分别检测其内Glu、GABA的含量。结果:与假手术组相比:模型组大鼠皮层Glu和GABA均升高,其中GABA更为显著(P<0.01);其余各组皮层的Glu、GABA则均略有降低,但均无统计学差异(P>0.05)。与模型组相比:安理申组、金纳多组以及参知健脑胶囊低、中、高剂量组鼠脑皮层Glu、GABA均有降低,其中参知健脑胶囊高剂量组的Glu和GABA、安理申组的GABA降低幅度更为明显(P<0.05)。参知健脑胶囊高剂量组大鼠皮层GABA降低较安理申组明显,但组间比较无统计学差异(P>0.05)。与假手术组相比:双侧颈总动脉永久结扎大鼠海马Glu和GABA略有升高或降低,但均无统计学差异(P>0.05)。与模型组比较:5个给药组2VO大鼠8周后海马Glu和GABA略有降低,但均无明显差异(P>0.05)。结论:参知健脑胶囊改善拟VD大鼠的学习记忆功能可能与降低皮层中的神经递质Glu和GABA的含量有关,但其具体作用机制尚需深入探讨。Objective:To observe the effect of Shenzhi Jiannao Capsule on Gln and GABA in cortex and hippocampus of vascular dementia rats. Methods : Seventy SD rats were randomly divided into sham - operated group, model group, Aricept group, Ginaton and Shenzhi Jiannao Capsule low, middle and high dose groups, 10 rats in each group. The 2VO method was used to establish rat vascular dementia model. Except sham - operated group and model group were given equal volume of normal saline, the other groups were given Aricept 1 mg/kg, Ginaton 20 mg/kg and Shenzhi Jiannao Capsule 40 mg/kg, 120 mg/kg and 360 mg/kg by gavage. 7 days after the operation ,administrations were given once a day and lasted for 8 weeks. After 8 weeks of administration, all the rats were executed to get cortex and hippocampus, and from which Glu and GABA were detected by HPLC method. Results: Compared to sham - operation group, Glu and GABA in cortex in model groups were both higher, particularly, GABA was more notable(P 〈 0.01 ). Glu and GABA in cortex of the other groups were slightly higher, but there was no statistical significant difference (P 〉 0.05 ). Compared to model group, Glu and GABA in cortex in Aricept group, Ginaton and Shenzhi Jiannao Capsule low, middle and high doses groups were both slightly lower, among which Glu and GABA in Shenzhi Jiannao Capsule high group and GABA in Aricept were even lower( P 〈 0. 05 ). GABA in cortex in Shenzhi Jiannao Capsule high group was lower than that in Aricept group,but there was no statistical difference(P 〉 0.05 ). Compared to sham -operation group, Glu and GABA in hippocampus in permanent bilateral common carotid artery occlusion rats were slightly higher or lower, but had no statistical difference( P 〉 0. 05 ). Compared to model group, Glu and GABA in hippocampus in all 5 medication groups were all slightly lower, but had no statistical difference (P 〉 0. 05 ). Conclusion:Shenzhi Jiannao Capsule could improve learning and memorial function of vascular dementia
