六味地黄汤早期干预对肥胖高血压大鼠的治疗作用及其机理研究

摘　　要：目的:观察六味地黄汤对肥胖高血压大鼠血压的影响,并从胰岛素抵抗及RAS两方面探讨相关机理。方法:40只SD大鼠分为正常组(n=10)及高盐高脂组(n=30)。高盐高脂组予饮食干预两周后分为模型组、中药组及缬沙坦组(均n=10)。每周测大鼠血压,第8周处死大鼠,称体重,检测血脂、血糖、胰岛素浓度,计算胰岛素抵抗指数(HOMA-IR),检测尿醛固酮、尿钾/钠比及血清血管紧张素Ⅱ(AngⅡ)浓度。结果:与正常组相比,模型组大鼠血压、体重明显升高,血脂异常,血糖及HOMA-IR升高;尿醛固酮亦明显升高。与模型组相比,中药组大鼠血压平稳下降;高密度脂蛋白升高;血糖显著降低;HOMA-IR明显下降;尿醛固酮及尿钾/钠明显降低。结论:中药六味地黄汤早期干预能有效降低肥胖高血压大鼠的血压,可能与有效提高胰岛素敏感性及抑制RAS激活有关。Objective ： To investigate the effects of Liuwei Dihuang Decoction on blood pressure of obese hypertension rats and its mechanism on insulin sensitivity and RAS. Methods ： Forty SD rats were divided into normal diet group（ n = 10） and high - salt and high -fat diet group （n = 30）, which were divided into model group, herb formula group and Valsartan group （n = 10）. After two - week diet intervention, blood pressures were measured every week. In week 8, weight of the rats was measured and blood plasma was collected to measure the lipid profile, blood glucose, insulin level and plasma Angiotensin Ⅱ level. HOMA - IR was then calculated. Urine aldosterone and Na/K were also measured. Results ： Compared to the control group, model group had higher blood pressure, body weight, blood glucose, HOMA - IR and abnormal lipid profile. The model group also had higher urine Na/K. Compared to model group, herb formula group＇s rats had lower blood pressure, higher high density lipoprotein, decreased blood glucose and HOMA -IR. Herb formula group also had a decreased urine aldosterone and Na/K ratio. Conclusion：Liuwei Dihuang Decoction can decrease the blood pressure of the obese hypertension rats,which maybe through both increasing insulin sensitivity and suppressing RAS.
