摘  要:目的:探讨复制COPD大鼠模型的方法并总结其经验。方法:从184只SD大鼠中随机选取18只纳入假手术组,其余166只用于造模。采用气道注射脂多糖加烟熏的方法复制COPD大鼠模型,假手术组于同一日气道注射等量生理盐水。观察大鼠一般情况及呼吸系统体征;光镜下观察大鼠肺组织;用ELISA法检测BALF中IL-1β、IL-6、IL-8、NF-κB、TNF-α炎性因子水平。结果:假手术组大鼠肺组织镜下见异常率为18.8%,模型组为92.1%;模型组大鼠BALF中IL-1β、IL-6、IL-8、NF-κB、TNF-α的含量与假手术组相比均有显著差异,提示造模成功,造模过程中死亡率为46.4%。结论:气道注射脂多糖加烟熏的方法可成功复制COPD大鼠模型,但尚需改善具体细节以降低死亡率。Objective: The measures of copying COPD rat model were investigated. Methods: 18 rats were selected and randomized into the sham-operated group; the remaining 166 rats were used for modeling and regarded as the model group. Rat model of COPD was copied by airway injection of lipopolysaccharide plus smoked way. On the same day, the same amount of saline was injected into rats in the sham-operated group. General situation and respiratory symptoms of rats were observed. Lung tissue of rats was observed under optical microscope. Levels of IL-1β, IL-6, IL-8, NF-κB, TNF-α inflammatory cytokines in BALF were detected by ELISA. Results: Abnormal ratio of rats lung tissue under optical microscope was 18.8% in the sham-operated group, and 92.1% in the model group. The difference of levels of IL-1β, IL-6, IL-8, NF-κB, TNF-α inflammatory cytokines in BALF between groups was statistically significant, prompted that the model was successfully established; Mortality was 46.4% during the modeling process. Conclusion: Airway injection of lipopolysaccharide plus smoked way could copy COPD rat model; hut specific details need to be improved for reducing mortality.
